In our previous works the cDNA clones BSD-84 encoding 84 kd human sperm protein and BED-20 encoding human testis cytosol protein were isolated and purified from human lambda gtll cDNA expression library. To determine their chromosome location, 1.35 kb BSD-84-BamHI and BED-20-BamHI cDNA fragments were used as probe to investigate the presence of the genes on human lymphocyte chromosome. Analysis of 3H-labeling autoradiography and together with Dig-labeling enzyme assay in situ hybridization to metaphase chromosome, resulted in the main experiment conclusions as follows:(1). 250 R-banding metaphase chromosomes were examined, and 1257 silver grains were scored, 22.3% of total silver grains was on chromosome 1 (p<0.005). About 40% of silver grains on chromosome 1 was located in the region lp35-36, this result was tallied with the results analyzed with Dig-labeling in situ hybridization and Southern blot hybridization with hybrid cell.(2). 87 R-banding and 56 non-banding lymphocyte metaphases were examined, 510 silver grains were scored, 11.56% of which is on human chromosome 14 (p<0.005) , about 5% of which is in the region 14q31-t.
