| Objective:(1)By sequencing and comparative analysis of urinary exosome mirnas from patients with Idiopathic Membranous Nephropathy(IMN)and healthy controls,differentially expressed mirnas were screened out.(2)Bioinformatics analysis of differentially expressed urinary exosome mirnas was performed to explore the possible pathogenesis of IMN,providing a new idea for clinical treatment of IMN.Method:(1)Urine samples were collected from 8 patients with IMN and 8 healthy controls in the nephrology department of The Third Bethune Hospital of Jilin University.(2)The expression profiles of two groups of exosome mi RNA were obtained by high throughput sequencing.Compared with healthy controls,patients with IMN outside secrete show significant difference in urine of micrornas with 73,of which 24 mi RNA can be retrieved in the mi RNA database.(3)The mi RNA expression profiles of the two groups of urine exosomes were obtained by high-throughput sequencing technology,and the differentially expressed mirnas were screened.(4)Bioinformatics was used to analyze GO and KEGG enrichment of differentially expressed mi RNA target genes.Results:(1)Urine exosomes from IMN group and healthy control group were extracted successfully.(2)The expression profiles of two groups of exosome mi RNA were obtained by high throughput sequencing.Compared with healthy controls,patients with IMN outside secrete show significant difference in urine of micrornas with 73,of which 24 mi RNA can be retrieved in the mi RNA database.(3)Among the 24 known differentially expressed mi RNAs,18 were down-regulated,and mi R-133a-3p and mi R-206 had significant differences.The expression of 6mi RNAs was up-regulated,and mi R-20b-5p had significant differences.(4)GO analysis showed that DNA-binding transcription activator activity and RNA polymerase-II specific were more significant in molecular function.Among the cellular components,presynapse was significant;In biological processes,differentially expressed mi RNAs are mainly involved in the regulation of trans-synaptic signaling and the modulation of chemical synaptic transmission.KEGG enrichment analysis showed that differentially expressed mi RNA target genes were mainly involved in MAPK,endocytosis,autophagy,Wnt,Notch and other signal transduction pathways.Conclusions:(1)This study showed significant differences in the expression of 24 known urinary exosome mirnas between IMN patients and healthy control subjects.(2)Among the 24 known differentially expressed mirnas,18 were down-regulated,among which mi R-133a-3p and mi R-206 had significant differences.The expression of 6 mirnas was up-regulated,and mi R-20b-5p had significant differences.(3)Differentially expressed mi RNAs are mainly involved in the regulation of trans-synaptic signaling and the modulation of chemical synaptic transmission.It is also possible to regulate the podococyte injury process of IMN by participating in various signal transduction pathways such as MAPK,endocytosis,autophagy,Wnt,Notch,etc. |
