| Objective: By studying the histopathological changes and the expression of epidermal barrier-related proteins in mice with psoriasis vulgaris,to explore the effect of Daihuang Xiaochuan cream on skin barrier function in mice with psoriasis vulgaris and its mechanism.Methods: Thirty-six male Balb/c mice were randomly divided into a blank group,a model group,Daihuang Xiaobi cream groups(low,medium and high concentration groups)and a carbotriol group after the removal of about 2×3 cm of dorsal villi after acclimatization.Except for Vaseline in the blank group,the other groups were smeared with 62.5mg/d imiquimod cream at 9: 00 am.every day for 7 days.At the same time,the model group and each treatment group were given the same amount of Vaseline cream,Daihuang Xiaobi cream and capotriol ointment at 17:00 in the afternoon for 7 days,and on the first,third,fifth and seventh day,the skin lesions of mice in each group were observed.The water content of skin lesions in each group was measured 24 hours after the last administration,and the skin lesions of each group were collected.The pathology and HE staining of mice in each group were observed,and the ceramides in the skin lesions were detected by ELISA.The expression of barrier related proteins mRNA of AQP3 CASP14 and FLG skin in the lesions were detected by q RT-PCR.Results: 1.The results of PASI score and HE staining showed that Daihuang Xiaobi cream could improve the erythema,scale,hypertrophy and pathological changes of psoriatic mice,and the middle concentration group had the best effect.2.Epidermal water content: The water content of Daihuangxiaobi cream in all groups was significantly higher than that in model group(P<0.05);Compared with low,high concentration and carpotriol groups,the water content of medium concentration group was higher than that of low and high concentration groups,but lower than that of carpotriol group(P<0.05).3.Epidermal thickness: The epidermal thickness of mice in Daihuangxiaobi cream group was significantly lower than that in model group(P<0.05);the epidermal thickness of medium concentration group was lower than that of low and high concentration groups and higher than that of carpotriol group,but the difference was statistically significant only between medium concentration group and low concentration group(P<0.05).4.CER content: The CER content of Daihuangxiaobi cream in all groups was significantly higher than that in model group(P<0.05);Ratio of medium concentration group to low,high concentration and carpotriol group,CER The content was higher than that in low and high concentration groups and lower than that in carpotriol group(P < 0.05).5.Skin barrier protein expression: The expressions of AQP3,CASP14 and FLG mRNA in Dihuangxiaobi medium dose group were significantly higher than those in model group(P<0.05);mRNA expression in medium concentration group was decreased compared with that in carpotriol group,but the difference was not statistically significant(P>0.05).Conclusion: Daihuang Xiaobi cream can improve the skin damage of mice with psoriasis vulgaris,restore the water content of epidermis,reduce the abnormal hyperplasia of epidermal thickness,and improve the skin barrier function of mice with psoriasis vulgaris.The mechanism may be that it increases the content of Cer and the expression of skin barrier protein mRNA such as AQP3,CASP14 and FLG in epidermis. |
