The plants of Hedychium genus(Zingiberaceae),especially their rhizomes part had edible and ornamental value.These plants also had good medicinal values.Studies showed that the chemical components of the Hedychium genus are rich,and some had good cytotoxic,antibacterial and anti-inflammatory biological activities.In order to find more and better chemicals with medicinal active ingredients in the plants of Hedychium genus,the chemical composition of Hedychium flavum plants collected from Dali,Yunnan was studied.The rhizomes of Hedychium flavum was crushed and soaked in methanol and extracted with ethyl acetate.Using a variety of separation techniques(CC and HPLC)and spectroscopic methods(IR,NMR and MS),49 compounds were isolated and identified from the ethyl acetate extract,which included twenty-seven diterpenoids(1-27),two sesquiterpenes(28 and 29),three monoterpenes(30-32),two indole derivatives(33 and 34),three simple aromatics(35-38),six diarylheptanes(39-45)and four steroids(46-49).Compounds 1-4 are new labdane diterpenoids and named as hedyflarins A-D,and the others are identified respectively as coronarin E(5),yunnancoronarin A(6),coronarin A(7),hedychenone(8),7-hydroxyl-hedychenone(9),coronarin D(10),hedychilactone D(11),calcaratarin D(12),yunnancoronarin B(13),coronarin D methyl ether(14),hedyforrestin B(15),villosin(16),hedyforrestin C(17),hedycoronarin C(18),isocoronarin D(19),curcucomosin C(20),hedyforrestin D(21),hedychilactone A(22),coronarin C(23),labda-8(17),13(14)-dien-15,16-olide(24),6-oxo-labda-7,11,13-triene-16-oic lactone(25),yunnancoronarin C(26),hedycoronarinB(27),acomadendrane-4β,10β-diol(28),1S,4S,5S,10R-4,10-guaianediol(29),(4R,6S)-6-hydroxypiperitone(30),terpin-3-en-1,5-diol(31),(3S,4S,6R)-p-menth-1-ene-3,6-diol(32),1H-indole-3-carboxadehyde(33),1H-indole-3-carboxylic acid(34),4-hydroxybenzaldehyde(35),4-hydroxy-3-methoxybenzaldehyde(36),3’,4’-dimethoxycinnamyl alcohol(37),bis-(3,4-dimethoxyphenyl)-methanone(38),(4E,6E)-1,7-bis-(4-hydroxy-3-methoxyphenyl)-hepta-4,6-dien-3-one(39),tsaokoarylone(40),gingerenoneA(41),5-hydroxy-1-(4’-hydroxyphenyl)-7-(4’’-hydroxy-3’’-methoxyphenyl)-3-heptanone(42),1,7-bis-(4-hydroxy-3-methoxyphenyl)heptane-3,5-diol(43),rel-(3R,5S)-3,5-dihydroxy-1-(4-hydroxy-3-methoxyphenyl)-7-(4-hydroxyphenyl)-heptane(44),coriandralpinin C(45),β-sitosterol(46),daucosterol(47),(22E)-5a,8a-epidioxyergosta-6,22-dien-3β-ol(48),and6-hydroxystigmast-4-en-3-one(49).Compounds 6-49 were isolated from Hedychium flavum for the first time.Structural modification from calcaratarin D(12),a large amounts of compound obtained from the plant,two derivatives villosin(16)and8,17-epoxycalcaratarin D(50)were obtained.Compound 50 was a new diterpenoid.Compounds 18 and 26 were tested for their inhibitory activity against triple negative breast cancer cells(MDA-MB-231 and HCC1806),with no associated activity.Compound 1-4,10,12,20,22,24-25 and 50 were tested for the cytotoxic activity of human breast cancer cells(MDA-MB-231)and human colon cancer cells(HCT-116),Only compound 10 showed some activity against the two cells,with the IC50values of 29.10μM and 23.43μM.
