| This article uses whey peptides and oyster peptides as materials,and uses wild-type Caenorhabditis elegans and human liver cancer cells as models to comprehensively evaluate the antioxidant regulation and hypoglycemic mechanism of short peptides.The results are as follows:1.Digested oyster peptides and whey peptidesα-The highest inhibition rates of glucosidase were 72.09%and 69.91%,respectively.The IC50 values of oyster peptide and whey peptide for hydroxyl radical clearance were 5.40 and 7.32 mg/ml,respectively,and the IC50 values of DPPH radical clearance were 6.66 and 2.53 mg/ml,respectively.The total reduction capacity was high,and both had good antioxidant activity.2.Oyster peptides and whey peptides can delay the increase of postprandial blood glucose in humans,resulting in a decrease in the peak postprandial blood glucose concentration,which is 8.63±0.15 mmol/L and 8.23±0.15 mmol/L,respectively.3.Whey peptides and oyster peptides can significantly prolong the lifespan of nematodes under normal growth conditions,high sugar environment growth conditions,and acute oxidative stress conditions,compared to the control group by 200%μG/ml concentration of whey peptides and 400μThe concentration of oyster peptides at g/ml prolonged by 19.65%and 28.13%,respectively(P<0.05).In nematodes,short peptides resist changes in high sugar environments by increasing the activity of antioxidant enzymes GPX and SOD(P<0.05).Whey peptides and oyster peptides can effectively reduce the content of MDA and ROS in nematodes under high sugar environments(P<0.05).4.The QRT-PCR results showed that whey peptides and oyster peptides increased the m RNA expression levels of genes such as daf-16,sir-2.1,and sod-3(P<0.05),altering the survival performance of nematodes in different environments.5.After establishing an insulin resistance model,the glucose consumption,glucose uptake,and glycogen content of cells decreased to varying degrees,with glucose consumption and glucose uptake being more significant(P>0.05).However,the treatment with short peptides restored the glucose consumption and glucose uptake ability of cells to some extent,and increased the glycogen content of cells.6.After establishing a cell oxidative stress model,the cell survival rate induced by H2O2significantly decreased(P>0.05)by 54.23%.The addition of short peptides protected the cells and reduced the damage caused by acute oxidative stress.Whey peptides and oyster peptides increased the cell survival rate to 69.77%and 76.87%,respectively.7.In a high glucose environment,by measuring the activities of CAT,SOD,and GSH-Px in cells,we found that both CAT and GSH-Px activities decreased and SOD activity significantly increased(P>0.05).Short peptide treatment can both enhance the CAT and GSH-Px activities of cells,with whey peptide being the most effective(P>0.05). |
