Due to long-term exposure to ultraviolet light,the skin becomes dry,rough,sagging,wrinkled and pigmented,and even leather-like lesions,and even skin cancer in severe cases.The main cause of photoaging is UV-driven oxidative stress,which leads to excessive production of free radicals in the body,causing oxidative damage to the skin.Rose essential oil(Reo)contains a lot of compounds such as terpenes,phenolic acids and flavonoids,which can scavenge free radicals and have a good antioxidant effect.However,Reo has poor water solubility,poor skin hydration when used directly,and a variety of active ingredients are volatile,resulting in short action time and low utilization.In order to increase the intensity and duration of the action of Reo locally on the skin,it is necessary to solve the above problems with the aid of new formulation techniques.Transethosomes(Tes)are composed of phospholipid bilayers,which are similar to skin lipids and can promote the passage of drugs through the stratum corneum;the addition of ethanol and surfactants makes them highly deformable and can increase the drug’s percutaneous penetration.In addition,Tes has a certain sustained release effect in the formation of drug depots in the dermis.This project intends to make Reo into rose essential oil alcohol transfer body(Reo-Tes),and then use cream as carrier to make Reo-Tes cream(Reo-Tes cream),the purpose is to use the bioadhesion of cream It can increase the retention time of Reo in the local skin,promote its percutaneous penetration,and realize the slow-release effect of Reo,in order to achieve better antioxidant effect,and treat and prevent skin photoaging.The main research contents are as follows:1.Preparation and characterization of physicochemical properties of Reo-TesThe content of Reo in Reo-Tes was determined by UV method and the methodology was investigated;Reo-Tes was prepared by ethanol injection method,and the drug loading and encapsulation efficiency were used as evaluation indicators,and the formulation composition and preparation process were optimized by single factor experiment;Reproducibility tests verify the feasibility and stability of formulations and processes.The results showed that the UV method for the determination of the Reo content in Reo-Tes was a simple method and met the requirements of the formulation process screening.The prepared Reo-Tes was spherical,with regular shape and no adhesion,and the particle size was(113.72±1.35)nm.The zeta potential was(-7.29±0.86)mv,the pH was(4.93±0.05);the EE and DL were(92.13±0.79)%and(15.80±0.44)%,respectively,stored at 40℃ for 30 days Good stability.2.In vitro release and antioxidant activity evaluation of Reo-TesThe content of Reo in pH 7.4 PBS(containing 1%Tween-80)was determined by UV method,and the methodological investigation results met the requirements.Taking Reo-Sol as the control and PBS with pH 7.4(containing 1%Tween-80)as the release medium,the in vitro drug release of Reo-Tes was investigated by dynamic membrane dialysis,and the mathematical model was fitted;The determination of three radical scavenging rates of O2-,·OH and DPPH verified the antioxidant properties of Reo-Tes.The results showed that the cumulative release percentages of Reo-sol and Reo-Tes in vitro at 2 h were(45.78±3.70)%and(23.21 ±2.90)%,respectively,and at 24 h were(79.18±3.13)%and(63.93±5.70)%,respectively;48 h were(79.55±2.99)%and(77.47±5.03)%,respectively.Compared with Reo-sol,Reo-Tes has a certain slow release,and its release curve conforms to the First-order kinetics equation;The scavenging rates of free radicals were 1.46,1.47,and 1.01 times that of Reo-sol,respectively,indicating that Reo-Tes did not affect the antioxidant capacity of Reo,but improved.3.Preparation and quality evaluation of Reo-Tes CreamReo-Tes cream was prepared by emulsification method.Taking appearance,spreadability,viscosity and centrifugal stability as evaluation indicators,the formulation and process of ReoTes cream were optimized by single factor test;Existence test and preliminary stability investigation;Franz diffusion cell method was used,with self-made Reo cream as the reference preparation,pH7.4 PBS(containing 1%Tween 80)as the release and permeation medium,the in vitro effects of Reo-Tes cream were tested.drug release(and transdermal permeability were studied.The results showed that the appearance of Reo-Tes cream was milky white,viscous and semi-solid,with good spreadability and skin feel,centrifugal stability was good;the cumulative release percentages of Reo cream and Reo-Tes cream in vitro were(20.83±1.65)%and(9.73±0.79)%at 2 h,and(69.50±4.49)%and(59.07±2.51)at 12 h,36 h were(69.77±4.72)%and(71.68±3.41)%,respectively,indicating that Reo-Tes cream has a certain sustained release;at 2 h,12 h and 24 h,the accumulation of Reo-Tes cream The transdermal penetration was 1.70,1.30 and 1.37 times that of Reo cream,which increased the transdermal penetration of Reo.4.In vivo anti-photoaging test and skin safety evaluation of Reo-Tes CreamFirst,the photoaging animal model was established by ultraviolet light irradiation,and then different drug preparations were given 30 minutes before ultraviolet light irradiation every day for protection.At the same time,the appearance of the skin was observed and photographed.After the experiment,the back skin of the mice was taken for histology.And biochemical indicators(SOD,CAT and MAD)were examined to explore the protective effect of Reo-Tes cream on mouse skin photooxidation;rabbit skin irritation and guinea pig skin sensitization tests were used to investigate the skin safety of Reo-Tes cream.The results show that the UV light irradiation method can successfully establish the photoaging model.Compared with the model group,the skin of the mice in the Reo-Tes Cream group was more delicate,smooth and elastic,with fewer wrinkles,which was close to the skin state of normal mice;the skin tissue level was clear,the collagen fibers were thick and tightly arranged,and there were new capillaries;biochemical indicators The activity of SOD and CAT increased,and the content of MAD decreased,indicating that Reo-Tes Cream has antiphotoaging effect.Compared with Reo Cream group,Reo-Tes Cream group has better antiphotoaging effect.Skin safety test results show that Reo-Tes Cream is non-irritating and sensitizing to the skin.In conclusion,Reo-Tes Cream was successfully prepared in this experiment and tested in vitro and in vivo.Reo-Tes Cream can promote the transdermal absorption of Reo and improve its anti-photoaging ability,and has a certain slow release effect,which is safe to use.It provides a new idea for clinical anti-skin photoaging research.
