| The bioactive components of Hericium erinaceus contain polysaccharides,peptides,fatty acyl substances and so on,and the most important ingredient is polysaccharide.H.erinaceus has good medical value,which can cure indigestion,gastric ulcer,gastritis,stomach pain,bloating,lowering blood glucose and so on.With the improvement of living standards,people’s consumption concept is changing,and the demand for beverages gradually changed from quenching thirst to healthy drinks.In recent years,Edible Fungi beverage is popular on abroad,but itis still difficult to see the healthy beverage products developed by H.erinaceus in the domestic market.Therefore,in this paper,H.erinaceus polysaccharide was extracted and developed into healthy beverage.The main results are as follows:1、In this paper,the traditional hot water extraction method,which did not degrade polysaccharides,was used to study the effects of extraction frequency,duration,temperature and material-liquid ratio on the yield of polysaccharide.Through orthogonal optimization,the best conditions for the hot water extraction of polysaccharide were that the extracting numbers and duration were 3 and 8h at 95℃,the ratio of solid material to liquid was 1:20.Under the optimal conditions,the crude polysaccharide extraction ratio was highest(8.30%).2、The effect of microwave on the yield of polysaccharide was studied.The optimum conditions of microwave method were obtained by single factor test and orthogonal test.The ratio of the solid material to liquid was 1:40,the extracting duration was 10min,and the microwave power was 140W.Under the optimal conditions,the extraction rate of polysaccharide was 5.51%.3、The effect of ultrasonic on the yield of polysaccharide was studied.The optimal extracting conditions for ultrasonic-water bath were obtained by single factor test and orthogonal test.The best conditions of ultrasonic method were that the ratio of solid material to liquid was 1:20 with extracting duration 20min at 50℃,the polysaccharide yield was up to 15.59%.Comparing the hot water extraction,ultrasonic extraction and microwave extraction method,the extraction rate of ultrasonic extraction method was the highest,which was 87.8%higher than the one of traditional hot water extraction.And ultrasonic extraction also had the advantages of simple operation,low cost,low solvent consumption and high extraction rate.4、The polysaccharide of H.erinaceus was extracted by optimal process parameters,then the beverage was prepared.The sensory qualities,microbial number,sugar content and heavy metals of the beverage were detected,respectively.The results showed that the formula was 1g aspartame,2g sucrose and 0.4g citric acid in the 500mL H.erinaceus extract,the taste was best.After 20 min sterilization at 105℃,the total bacterial number and coliform group meet the hygienic requirements.The contents of arsenic,lead,cadmium and mercury were very low,and conform to the GB7101-2015 standard.5、The health function of H.erinaceus beverage was preliminarily tested.The results showed that H.erinaceus beverage could inhibit the growths of harmful Staphylococcus aureus and Salmonella enteritidis.The effects on Streptococcus thermophilus and Lactobacillus bulgaricus growth were not significant.The results of this study laid a foundation for further development of H.erinaceus beverage. |
