Maize is one of the world’s most important food crops.It originated in southern Mexico from the domestication of the ruminant grass and is a photoperiod sensitive crop.As an important trait in maize fertility,the targeted regulation of maize photoperiod has become a hot topic in maize trait improvement.In previous studies,it has been shown that maize flowering is regulated by a combination of genes and components in a very complex response mechanism,and that among these genes-ZCN8 is an important gene that has been shown to be involved in the maize flowering regulatory pathway.In this experiment,12 targets were designed for gene editing in the promoter region of the ZCN8 gene,with the aim of selecting the editing sites that would have the most impact on flowering in maize.The efficiency of multi-fluorescent co-hybrid Agrobacterium-mediated genetic transformation used in this study was also evaluated to measure the feasibility of multi-vector transformation implementation.The results obtained in this study are as follows:1.Multiple fluorescent markers of multiple gene editing vectors can be mixed to obtain more transformants and a richer variety of gene editing types by reducing the number of genetic transformations.2.Precise regulation of flowering by fine-tuning the ZCN8 gene through promoter editing is feasible and effective,and three late-flowering genotypes were obtained through promoter editing.3.The Sp1 photosensitive element plays an important role in the regulation of flowering and is presumed to be an important element associated with flowering.4.The absence of the relevant components of erythromycin synthesis can seriously affect plant height under long day conditions.
