| Aeromonas hydrophila is a gram-negative bacterium.It is widely distributed in nature and is commonly found in fresh water,sewage,silt,soil and human feces.It is the main pathogen that causes Aeromonas hydrophila sepsis in fish,with a The mortality rate is extremely high.A.hydrophila is one of the most powerful pathogens in the aquaculture industry in my country,a long epidemic,and huge economic losses.The use of antibacterial drugs to treat diseases related to Aeromonas hydrophila has significant effects,but its harmfulness is also very prominent.Antibiotics and chemical drugs will not only form harmful residues in the fish.It can also increase the resistance of bacteria and spread the resistance widely.Therefore,the development of a new type of green,non-residual,non-toxic and side-effect medicament is of great significance to the prevention and treatment of aquatic products.Three lactic acid bacteria were selected in this experiment,namely Lactobacillus rhamnosus,Lactobacillus casei and Lactobacillu plantarum.Fermentation is carried out through the acid production characteristics of lactic acid bacteria combined with the medicinal effect of the Chinese medicine Sanyu,which further stimulates the active ingredients in Sanyu,and enhances the medicinal effect.Furthermore,A.hydrophila was subjected to in vitro inhibition test,inhibition biofilm detection and enzyme activity analysis.The results showed that the fermentation products obtained from LGG fermentation of Sanyu,LC fermentation and LP fermentation of Sanyu all have inhibitory effects on A.hydrophila.Among them,the fermentation product of LGG+Sanyu has the strongest inhibitory effect on A.hydrophila,followed by LC+Sanyu and LP+Sanyu.In addition,LGG fermented Sanyu products also have a certain inhibitory effect on the biofilm formation ability of A.hydrophila.Therefore,in general,LGG+Sanyu has the most obvious inhibitory effect on A.hydrophila.The enzyme activity analysis results of the three fermentation products showed the presence of protease,lecithinase and urease in the product,which can enhance the organism metabolic function.In order to assess the immune effect of the fermentation of Chinese medicine in lactic acid bacteria,this test is used as an immune-like subject,and three fermentation product packages are fed the test,and the feed of 56 d is fed continuously,and the immunized 14 d,28d,42 d and 56 d Serum and tissue were collected by 56 d to detect the level of the corresponding immune indicators.The end weight and calculated endors were measured after 56 days.Immunoglobulin Ig M,complement C3 and C4 were detected by ELISA.Differential fungal fungus can significantly improve blood veraffin immune indicators,where LGG fermentation,eliminated,Ig M,C3 and C4 levels significantly higher than LC fermentation territory and LP fermentation.By detection of antioxidant related enzymes CAT,SOD and GSH-PX in serum,it was found that the fermentation of lactic acid bacteria fermentation can significantly improve the antioxidant enzyme activity in the serum,which is consistent with the immune-related indicators,and the LGG fermentation optimal.The expression level of immune-related genes in each group was detected by q RT-PCR.As a result,it was found that the fermentation of lactic acid bacteria can significantly increase the level of IL-1β,IL-10,TNF-α and IFN-γ in the intestine,wherein the LGG fermentation density can significantly increase the liver,spleen and kidney IL-10,IL-1β.And the expression level of TNF-α,My D88 as a joint molecular protein of the Toll-like receptor family,can promote cellular immunity.The expression level of the My D88 gene in the liver,spleen,kidney and intestines in LGG,further indicate that LGG fermentation demon can increase the immune level of host cells.After the end of the feeding trial,2d was stopped,and the abdominal injection A.hydrophila was performed,14 days was observed,and the results found that the immunological protection rate of LGG fermentation Chinese medicine is 60.87%,LC fermentation territory and LP fermentation Division It was 56.09% and 41.46%,indicating that the LGG fermentation ground protection effect was better than LC fermentation territory and LP fermentation.It is further proved that LGG fermentation demon can significantly increase the ability of the A.hydrophila invasion.In summary: This test passes through in vitro tests,verifying the fermentation product of three lactic acid bacteria can effectively inhibit A.hydrophila growth and generating film,prove that the fermentation product of three lactic acid bacteria can effectively activate the host non-specific immune system by in vivo trials.Immune correlation indicators,while improving the immunity of antioxidant enzyme activity in serum.The results of this study provide theoretical basis for the development and application of new green non-toxic side effects. |
