Obtaining And Identification Of Distant Hybridization Progeny Of Brassica Napus And Capsella Bursa-pastoris

In order to improve the germplasm resources of innovative Brassica napus and make the excellent characters unique to other species become the focus of traditional breeding.Herba shepherd’s purse and brassica napus with medicinal value were selected for distant hybridization to obtain new germplasm resources and improve the medicinal value of rape.So the far-flung hybrid innovation breeding of rape is the hot spot of current breeding.Shepherd’s purse and rape are the species of cruciferous family,but they are of different genera,so the relationship is far away,and the difficulty of distant hybridization is higher.In order to improve the success rate of distant hybridization between rape and shepherd’s purse,this paper carried out distant hybridization experiments with different varieties of rape and shepherd’s purse,respectively,to obtain excellent distant hybridization combinations,and to identify and analyze the obtained seeds.1.Different varieties of rape and shepherd’s purse were crossed separately,and the optimum combination of NARC and shepherd’s purse was selected by the parameters of angle rate and affinity index obtained.The angle ratio and affinity index of the stigma treated with 1% sodium chloride solution were the highest.2.The analysis of different fatty acid content of seeds of rape and shepherd’s purse by gas chromatograph,it was found that the soft fatty acid content of hybrid offspring showed an increasing trend compared with that of parents,while the content of oleic acid,linoleic acid and linolenic acid showed a decreasing trend,and the content of erucic acid also increased compared with that of.3.Observed the pollen grain morphology of the F1 generation by microscope,and found that the pollen grain morphology of the hybrid offspring was irregular,and the pollen was flat and small.The horn fruit and morphology of the hybrid progeny also showed irregular morphology,the seed distribution in the horn fruit was uneven,the number of seeds was small,the seed shape was irregular,the surface was uneven,most of the colors were dark reddish brown or black,and a few showed yellow seed.4.Through distant hybridization to obtain an sterile plant,stamen without pollen production,stigma growth protruding buds,between petals are not compact and wilting,flowers are smaller than the mother flower.The other hybrid progeny have obvious differences with their parents.Branching position of F1 hybrid progeny changed,the degree of stem lignification decreased,stem thin and easy to bend,the number of branches and the number of horned fruit,the number of seeds are lower than the mother.5.some 75% of the genes were from the rape genome and 20% from the shepherd’s purse genome,it can be concluded that the hybrid progeny of these four different varieties and shepherd’s purse is indeed the interspecific hybrid progeny of the two species.Among them,11 y802 crossed with shepherd’s purse had the highest genome containing shepherd’s purse,reaching 20.18%.
