Red Pear is favored by people,with a broad market value.The color of red pear mainly depends on the synthesis and accumulation of anthocyanin,and there are many factors affecting the biosynthesis of anthocyanin,among which light is one of the most important factors.There are few studies on the effect of light on pear anthocyanin,and there is a lack of systematic study on the mechanism of the effect of light on anthocyanin.Therefore the experiment to the light of the different processing Pingguoli,carries on the determination of pigment content and the use of a new generation of high-throughput sequencing technologies for the transcriptome analysis,screening differentially expressed genes related with red fruit development,to further study the illumination effect on pear flower green glycosides,the main results were as follows:1.Determine the anthocyanin content and coloring index of Pingguoli treated with natural light,daylight shading and night light.The results showed that the fruit of natural light treatment was completely colored after unbagging,and the coloring index was 0.75.The color index of the shading fruit was 0.25 in the daytime,and the color of the shading fruit was abnormal at night,with many red spots on the fruit surface.The content of anthocyanin in pericarp treated with natural light was the highest,while the content of anthocyanin in pericarp treated with shading in daytime was the lowest,and the content of anthocyanin in pericarp treated with night light was in the middle.2.Transcriptome sequencing was completed in this analysis,and a total of 58.62 G Clean Data was obtained.The available data volume of each sample ranged from5.96 to 6.84 G,the Q30 base distribution ranged from 95.47 to 95.72 %,and the average GC content was 47.78%.24447 pieces of Unigene were splice together,with a total length of 43070,268 bp and an average length of 1276.61 bp.Reads were compared with Unigene,and the comparison rate was 86.1-86.87%.There were 3differential groups.According to CK_VS_ZD comparison,2400 differential genes were expressed,among which 1138 genes were up-regulated and 1262 genes were down-regulated.CK_VS_ZG showed 4825 differentially expressed genes,among which 2591 differentially expressed genes were up-regulated and 2234 differentially expressed genes were down-regulated.Compared with ZD_VS_ZG,there were 2298 differential expressions,including 977 up-regulated and 1321 down-regulated differential expressions.3.Fifty different genes were screened and related pathway analysis was described,and it was found that MYB4-like1 gene was in the pathway of gene information processing,which further indicated that MYB4-like1 gene may be a key regulatory gene that changes under light regulation and then affects the content of anthocyanin.Moreover,it was found that GLT1 gene,HY5 gene,4CL gene and GST gene participated in the related pathway of anthocyanin synthesis,which laid a foundation for further experimental verification.4.PCR identification of the different genes under different light treatments revealed that genes such as MYB4-like1 and BHLH130-like were significantly up-regulated,and the differential expression of WDR gene was significantly the same as the content of anthocyanin.
