| Water lily is a famous ornamental plant,but the the vase life of its cut flower is limited,and it is easy to produce bent stem during vasing life.In this study,Nymphaea’Blue Bird’was used as the experimental material to study some physiological indicators and the ornamental characters during vasing life.Different treatments were carried out to search the best method to prolong the vase life of waterlily.At the same time,transcriptome analysis,hormone content detection and paraffin section observation were performed on the curvilineous stem of water lily.The main research results are as follows:(1)In nature,the flowering duration of waterlily is about 3 days.Within one day,the maximum value of the flower diameter appeared at about 11:00 noon every day,the stem gradually bended during flowering period.After use water bottle inserts,the stem of water lily is obviously bent on the first day,on the fifth day,the flowers closed,the results showed that the curving stem was one of the key factors affecting the preservation of waterlily cut flowers.(2)A3B3C3D2(A3:Base preservative of 10 g starch+5 g sodium bicarbonate,B3:Add 50 mg/L gibberellin to the basic preservative,C3:calcium nitrate of 5mmol/L,D2:40mg/L gibberellin is added to the liquid bottle inserts)treatment can extend the vase life of water lily cut flowers.The experimental results showed that the treatment could significantly increase the flower diameter,delay the decrease of the protein concentration and CAT activity,slow down the increase of the relative conductivity.(3)Transcriptome data results showed that 607 differentially expressed genes were identified in the lotus bendy stem,of which 247 were up-regulated and 360 were down-regulated.There were differences in the expression of genes related to plant hormone synthesis pathway,gravity,starch granules and Ca2+photosynthesis.The results were verified by q PCR and were consistent with the transcriptome sequencing results.(4)The contents of abscisic acid(ABA),indole-3-formaldehyde(ICA),jasmonic acid-isoleucine(JA-ILE),jasmonic acid(JA)were significantly different.Salicylic acid(SA),dihydrogen zeatin(DZ),cis zeatin(c Z),trans zeatin(t Z),pentene adenine(IP),and indole-3-butyric acid(IBA),indole-3-methyl acetate(ME-IAA),indole-3-acetic acid(IAA)showed little difference in the stem of water lily.(5)During vasing life,the morphology of dorsal and abdominal cells has changed.The direction of settlement is consistent with the bending direction of the water lily stem,as well as the direction of gravity.This research provided an effective method to increase the vase life of water lily,and analyzed the possible theory of the stem bending during the vase life of waterlily. |
