Panax ginseng C.A.Meyer is known as the "King of Herbs".1-5 years old ginseng can be used as food As a medicine and food homologous plant,it is widely used in food and health products industry.Ginsenoside has many pharmacological activities and is a marker compound for evaluating the quality of ginseng.Jilin Province is the main area to produce ginseng in China,furthermore,the Yanbian Korean Autonomous Prefecture(Yanbian)is one of the major areas to produce ginseng in Jilin Province.However,the mechanism of ginsenoside accumulation in Yanbian area has not been reported yet.In this study,the cultivated ginseng of the one,two and three years in Yanbian area were as the test materials to study the change of ginsenoside accumulation and the expression characteristics of the key enzyme gene(PgDAS)in the ginsenoside biosynthesis pathway during its growth;at the same time,the effect of temperature on the accumulation of ginsenosides was analyzed,which provided a scientific theoretical basis for the cultivation of ginseng in this area.The main results of this study are as follows:The accumulation of ginsenosides was tissue-specific,and the total ginsenoside content in leaves was higher than that in roots and stems.At the same time,the content of underground parts gradually increased with the increase of plant age,while the aboveground parts gradually decreased;During the growth of ginseng,the content of protopanaxadiol-type(PPD-type)ginsenosides were leaf>root>stem in different tissues,while the protopanaxatriol-type(PPT-type)were leaf>stem>root;The contents of ginsenosides Rb1,Re,and Re were the highest in ginseng roots,stems,and leaves,respectively.The harvest period significantly affected the accumulation of ginsenosides.The ginsenoside content and biomass of the one,two and three years were higher in July and August,it shows that the appropriate high temperature is conducive to the accumulation of ginsenosides.Real-time quantitative PCR shows that PgDAS gene is constitutively expressed in different tissues and has tissue specificity.The expression level was leaf>root>stem.At the same time,the expression level in three years was significantly higher than that in the one and two years;overall positive correlation with total ginsenoside accumulation.
