| The paper studied the explore structure and physicochemical properties of new substances produced by the reaction between phenolic compounds and IAA under the catalysis of polyphenol oxidase(PPO).It also regards Eucalyptus plantlets ‘Eucalyptus grandis’ as tested materials,to investigate the reaction product of Eucalyptus plantlets rooting effects,to observe and get statistical rooting.Simultaneously,to determinate the related enzymes of adding reactants during the test tube plantlets rooting process,such as peroxidase(POD),polyphenol oxidase(PPO)and indoleacetic acidoxidase(IAAO)and endoggenous hormones.Exploring the relationship between the reaction product and adventitious roots of eucalyptus shoots to provide theoretical guidance for solving the problem rooting of test tube seedlings of woody plants.The main findings are as follows:1.The products were separated and purified by HPLC.We found that the purity of the product was high,and meet the qualification requirements.At the same time,we carried out molecular weight identification of the product by UPLC-Q-TOF-MS.The results showed that the product was small molecular compound.2.Eucalyptus of test tube plantlets ’ optimum rooting culture was 1/2MS + NAA0.5mg L-1 +IAA 0.2mg L-1+ Plant gel 2g L-1,the rooting rate was 61.7%.While on this rooting medium supplemented with 0.2mg L-1 product can increase the rooting rate up to 77.8%.IAA and the products showed that the rooting of test tube seedlings of eucalyptus were promoted.The addition product was higher than that of IAA in rooting rate,maximum root length and root length.But the two treatments were not significant differences in root activity.3.By measuring eucalyptus test tube seedlings on the 20 th day of soluble sugar and soluble protein content in different treatments and found that differences in the results of the three groups under treatment level are not significant.4.Study adding products on Eucalyptus ’grandis’ POD,PPO,IAAO activity dynamic changes during the rooting process.The results show that: compared with the C1,after adding the product(C2),Eucalyptus plantlets of POD activity peak in the whole growth process appeared twice,respectively in the 2d and 7d,and C1 pod peak only once on 4d,but differences in overall content was not significant.The content of PPO in the C2 treatment level showed a rising trend,and the differences was significant compared with C1;From the root induction phase to elongating stage(2d-7d),the PPO content of C2 was increased,which was higher than that of PPO in the same time period C1.The IAAO enzyme activity of the three treatments showed an upward trend,and the change trend was basically the same,but the IAAO activity in the C2 treatment level was higher than that of C1 during the whole growth process.5.compared to the three treatment of endogenous hormone ZT,IAA,ABA,GA3 content changes,we found that the treatment of C2 after the addition of the product IAA,ABA the overall content level is higher than C1,while the ZT,GA3 content is lower than the overall level of C1. |
