| This experiment on cucumber pollen mother cell meiosis,karyotype analysis,pollen development and pollen culture four aspects are studied,aims to provide some reliable data for cucumber breeding work.Its main contents are as follows:1、Studied on meiotic behaviors of pollen mother cells in cucumber.The result suggested that there is a certain degree of desynchronization in the courses of meiotic.It displayed in two aspects:1)in the same anther of pollen mother cells,all sorts of different periods cells were observed from metaphase Ⅰ to telopphase Ⅰ;2)The division of chromosome also was not synchronization in the same anther.A dyad chromosome had separated from the anaphase Ⅱ,but,the other not.2、Studied on the chromosome number and karyotype analysis of cucumber.The result suggested that the chromosome number of cucumber was 2n=14,the chromosome base was 7,the chromosome karyotype formula was 2n=14=14m.Asymmetype index(As.k%)was 56.25%.The karyotype was 1 A and belonged to symmetrial type.Cucumber might belong to original species in phylogeny.3、Using the buds of cucumbers,which were stained with aceto-carmine dying solution,as meterial the pollen developments were obsorved with light microscope and the relationship between the bud size and pollen development stages were discussed.The results showed that the pollen was basically at the stage of microsporocyte and the peak stage of dyad when the size was 0-0.3 cm.While,tetrad,uninucleate、binucleate and trinuclear stage when the bud size were between 0.3-0.6 cm,and late-pollen development of pollens appeared the bud size were above 0.6 cm.4、At the pollen culture conditions described in this study,add the 0.5μm of TSA(Trichostain A,the agent of histone deacetylase inhibitor)to the culture medium effectively enhance the frequencies of cell division of pollen,all of the cucumber varieties used in the study expression cell division in the pollen. |
