| Poly(4-hydroxybutyrate)(P4HB),as a bioabsorbable material,can be used as a dura substitute in organisms.It not only has good mechanical properties,cellular compatibility and biocompatibility,but also does not accumulate acid degradation products in human body when degraded.Currently,commercial P4 HB is generally produced by bacterial fermentation,but it is difficult to be applied in the cerebrospinal system due to the high limitation of intracellular endotoxin.Therefore,P4 HB prepared by ring-opening polymerization of γ-butylactone has very high application value.This paper focuses on the synthesis of P4 HB fiber membrane and its application in vivo.1.P4 HB electrospinning fiber film was prepared and synthesized by electrostatic spinning technology,and determined by nuclear magnetic spectrum.2.The mechanical strength,contact Angle,GPC and TGA of P4 HB fiber membrane were measured.The above determination proved that the mechanical strength and decomposition temperature of P4 HB fiber membrane were sufficient to match the natural meninges.3.The cytotoxicity,adhesion and proliferation of P4 HB fiber membrane were measured by cell experiments.It was proved that P4 HB fiber membrane has the ability of inducing rapid cell migration,adhesion and proliferation of fibroblasts in vitro.4.The P4 HB membrane was implanted subcutaneously in rats,which proved that it had good biocompatibility and suitable biodegradation behavior.It was found that P4 HB membrane can prevent cerebrospinal fluid leakage and regeneration of dura tissues without local or overall infection and foreign body reaction.The successful animal experiments proved that P4 HB membrane is expected to replace natural meninges and be applied in clinical practice. |
