The earliest microscopes can date back to the late 16th century and were made by a Dutch cobbler.Since its inception,improvements in microscopy have never been abandoned,and microscopy technology has been carried out in various research fields by constantly improving its resolution,operability and applications.This thesis is divided into two parts including the characterization of wool keratin patterns using various microscopic techniques and the characterization of the catalytic behavior of vanadium pentoxide nano-enzymes using total internal reflection super-resolution fluorescent microscopy,with the following main research contents and conclusions.(1)A well-dispersed solution of wool keratin was successfully extracted using the alkaline method without the surfactant sodium dodecyl sulfate.Then we modified the wool keratin to make it photosensitive without significantly altering its structure and function which resulted in the synthesis of a green and environmentally friendly photoresist,and from which a large area of protein patterns were subsequently successfully prepared.Finally,the keratin patterns were found to be complete,highly uniform,and accurate characterized by scanning electron microscopy,3D confocal microscopy,and atomic force microscopy.(2)Vanadium pentoxide nanorods of different lengths were prepared by adjusting the synthesis parameters and their peroxidase activity was verified.To investigate the detail information concealed by the mean detection method,the catalytic behavior of vanadium pentoxide nanoparticles was characterized and super-resolution images were reconstructed with a home-built total internal reflection super-resolution fluorescenct microscope,using Amplex Red as a fluorescence probe in combination with the advanced algorithm a-live PALM single molecule localization technology.
