In our previous study,we found that the community structure of endophytic fungi in Dendrobium candidum produced in different localities had regional differences.Based on this,we proposed a new way to identify the origin of Chinese medicinal materials based on the regional differences of endophytic fungi community structure.In order to further verify the feasibility of this method,the construction process of endophytic fungi community structure in Dendrobium candidum during artificial cultivation and the regional differences of endophytic fungi community structure in Dendrobium candidum from Zhejiang,Sichuan and Yunnan provinces were systematically studied,and the correlation between the regional differences of environmental microorganisms in different producing areas was discussed.The formation mechanism of regional differences in endophytic fungal community structure of Dendrobium candidum was studied.Through metagenomic sequencing of Dendrobium candidum samples from Zhejiang,Sichuan and Yunnan,abundant information of endophytic fungi community structure was obtained,and the community structure characteristics of different producing areas were compared in different time and different space.The existence of regional differences was systematically verified,and the regional differences could be effectively distinguished among producing areas.The preliminary screening of endophytic fungi strains with ground-specific distribution laid a foundation for molecular labeling of Dendrobium candidum.By comparing the metagenomic sequencing of fungal community structure in different localities,it was confirmed that the fungal community structure in different localities of Dendrobium candidum had obvious regional differences in cultivation matrix,air and irrigation water.Some fungi only appeared in specific habitats.The correlation study showed that the structure of endophytic fungi community in Dendrobium candidum was closely related to the fungi community structure in the environment,especially the fungi community structure in cultivation medium and irrigation water had a greater impact on the structure of endophytic fungal community.In this study,high-throughput sequencing technology was used to obtain the population distribution and community structure information of endophytic fungi in Dendrobium candidum and fungi in environment from different localities,and theinfluence of fungi in environment on the community structure of endophytic fungi in Dendrobium candidum was discussed for the first time,which enriched the community structure information of endophytic fungi and confirmed that the regional differences in the community structure of endophytic fungi in Dendrobium candidum originated from different production.The differences of geo-environmental conditions can be used as environmental labels in the identification of Dendrobium candidum.
