| [Background and Objective]Tetramethylpyrazine(TMP)is a kind of alkaloid extracted from traditional Chinese medicine with blood circulation,anti-oxidation and anti-inflammatory.It protects the kidney and stabilizes kidney function.The damage of glomerular podocytes is the pathological basis of various nephropathy injuries.The PI3K/AKT pathway has been found to play an important role in the development and outcome of kidney disease.This study will establish a podocyte model of puromycin damage,and explore whether TMP can activate PI3K/AKT signaling pathway to antagonize the damage of puromycin to podocytes in vitro.[Methods]1.Establish a podocyte injury model by inducing podocyte injury by puromycin.The optimal model concentration of puromycin was screened by MTT assay for podocyte viability.2.To explore the protective effect of TMP on puromycin-induced podocyte injury model.MTT assay for podocyte viability to screen for theoptimal therapeutic concentration of TMP and to observe its protective effect.3.To investigate the effect of TMP on the PI3K/AKT pathway in the puromycin-induced podocyte injury model.The expression of nephrin protein,p-akt and akt was detected by Western Blotting.The activation of intracellular PI3K/AKT pathway in TMP-induced podocyte model was observed.4.To investigate the effect of PI3K/AKT pathway inhibitor LY294002 on the protective effect of TMP in puromycin-induced podocyte injury model.The expression of nephrin protein,p-akt and akt was detected by Western blotting to observe the inhibitory effect of PI3K/AKT pathway.[Results]1.Puromycin can induce podocyte injury to decrease cell viability and down-regulate the expression of nephrin protein.2.TMP can improve the cell viability of the puromycin-induced podocyte injury model and up-regulate the expression of nephrin protein.3.TMP can activate the puromycin-induced podocyte injury model PI3K/AKT pathway and regulate the expression of p-akt.4.PI3K/AKT pathway inhibitor LY294002 canreverse the improvement of TMP on the cell viability of podomycin-induced podocyte injury model and down-regulate the expression of nephrin protein.[Conclusion]Tetramethylpyrazine activates PI3K/AKT signaling pathway to protect puromycin-induced podocyte injury model... |
