| TRPV1(transient receptor potential cation channel subfamily V member 1)and TRPV2(transient receptor potential cation channel subfamily V member 2)are important channels involved in pain signal transmission in the dorsal root ganglion(DRG)cell body,channel activity Changes affect the degree of pain tolerance.It is understood that there is a difference in pain tolerance between goats and sheep,that goats are poorly tolerated,and that sheep are more tolerant to pain.In order to further clarify the causes of the differences in pain tolerance between goats and sheep,this paper studied the effects of differentially expressed genes of goat and sheep and blank control group on the expression and activity of TRPV1 and TRPV2 after Fos’s adjuvant inflammation and pain treatment.Goats and sheep respond to pain-regulating pathways.Combined with the mRNA and protein structure differences of TRPV1 and TRPV2 genes in goats and sheep,the causes of differences in pain tolerance between goats and sheep were determined.The results are as follows:(1)The goat mainly activates the TRPV2 channel when it is subjected to noxious stimulation.The sheep mainly activates the TRPV1 channel in response to pain.From the gene expression level analysis,the TRPV2 gene level and protein level expression of goat and sheep are consistent.(2)The goat TRPV1 gene transcript 1(TRPV1 X1)is identical to the TRPV1 gene transcript gene level and protein level in sheep;the sheep TRPV1 gene transcript is 180 bp more than the goat TRPV1 gene transcript 2(TRPV1 X2).(3)The protein structure of goat TRPV1 X2 is loose relative to the structure of sheep TRV1;goats and sheep have different regulatory pathways involved in the response to pain.Goats primarily affect TRPV2 channel activity in pain,while sheep primarily affect TRPV1 channel activity.Combined with the above results,it is concluded that the mRNA sequence and structural differences of TRPV1 in goats and sheep are one of the reasons for the difference in regulation and tolerance of goats and sheep in response to pain.Among them,goats and sheep have long-term differential expression of TRPV1 mRNA sequence,which is the main functional site affecting the pain tolerance of goats and sheep.This functional site will provide a theoretical basis for the research and development of new analgesic drugs. |
