| Actinidia arguta is the king of fruits of a new generation,after years of artificial cultivation and development has become a common food in people’s lives everyday.Actinidia arguta compared to other fruits rich in both nutrients,but also has an effective biologically active substances.This article provides Actinidia arguta alkaloids extraction technology,as well as separate issues antioxidant activity of the active ingredients to provide a scientific basis,the following studies:In this experiment,using response surface methodology extraction process Actinidia arguta alkaloids optimized using Design-Expert8.0 test and analysis software.The test results are as follows:The optimal extraction conditions for the alkaloid model in soft kiwifruit pulp were:methanol concentration of 75%,liquid-to-liquid ratio of 11.89:1,ultrasonic power of 250 W,and ultrasonic time of 10 min.Under this condition,soft jujube kiwifruit The extraction rate of alkaloids in the pulp was 0.572 mg/g.In combination with the actual conditions,the verification test conditions were set to: methanol concentration of 75%,liquid-to-liquid ratio of 12:1,ultrasonic power of 250 W,and ultrasonic time of 10 min.The extraction quality of soft acacia kiwi fruit alkaloids was 0.571mg/g,which was basically consistent with the predicted conditions,which proved the feasibility of the model.The optimal extraction conditions for the alkaloid model in the kiwifruit peel of soft jujube were: methanol concentration of 74.57%,liquid-to-liquid ratio of 19.50:1,ultrasonic power of 210.98 W,and ultrasonic time of 17.76 min.Under this condition,soft The extraction rate of alkaloids from jujube kiwi fruit peel was 0.760 mg/g.In combination with the actual conditions,the verification test conditions were set as follows: methanol concentration was 75%,liquid-to-liquid ratio was 20:1,ultrasonic power was 210 W,ultrasonic time was 18 min,and the extraction quality of soft apricot kiwi peel alkaloid was0.758 mg/g and prediction.The conditions are basically consistent,which proves the feasibility of the model.In this experiment,the alkaloids in the soft jujube kiwifruit were separated and purified by fractional extraction method to obtain the alkaloid-containing components,and themacroporous resin was separated.In this experiment,D-101 macroporous adsorption resin was used to purify the alkaloids in soft jujube kiwifruit.Ethanol was selected as the eluent,and 75%,85%,and 95% ethanol could elute the alkaloids.When the ethanol concentration of the eluent reached 75%,the alkaloids eluted more.The effluent was subjected to wavelength scanning(190 nm to 600 nm),and most of the peaks of the ultraviolet peak scan containing alkaloids were concentrated at 197,213,241,247,343,355,376,and 459 nm.The alkaloids are continuously eluted with the sample,and the eluate is subjected to thin layer chromatography to develop color.The color reaction is in the late stage of the eluent,and the ethanol concentration of most eluents is greater than 75%.The color reaction confirmed that an alkaloid compound was indeed present in the eluate.In this experiment,high performance liquid chromatography was used to separate and purify alkaloids from soft jujube kiwifruit with good results.Mobile phase: A is methanol and B is ammonia(p H = 10).The flow rate is 3m L/min,the injection volume is 2μL,and the detection wavelength is 250 nm.The composition of the alkaloids of Actinidia chinensis var.chinensis is 6 kinds,and the retention time is 5.450,6.446,6.956,11.370,11.698,21.147 min.In this experiment,the biological activity of antioxidant activity of total alkaloids extracted from the pulp and peel of soft jujube kiwifruit was determined and compared.The indicators of antioxidant activity include superoxide anion radical,hydroxyl radical,DPPH.Free radicals,reducing power.The results were as follows: the total alkaloids in the fleshy and peel skin of soft jujube increased the superoxide radical scavenging ability with the increase of concentration,which was positively correlated.When the mass concentration of kiwifruit alkaloids was more than 200μg/m L,the scavenging rate of superoxide anion radicals was saturated;in the range of 50-200μg/m L,the scavenging effect and extraction of hydroxyl alkaloids from the total alkaloids in the pulp and peel of soft jujube macaques.The mass concentration of the substance is positively correlated,that is,the clearance rate increases with the increase of the mass concentration of the alkaloid compound of the soft kiwifruit.The higher the concentration,the better the antioxidant effect;in the range of mass concentration 50-200μg/m L.The scavenging rate of DPPH free radicals of kiwifruit alkaloids increased with the increase of the concentration of the sample solution,which was almost the same as that of the control group,and the scavenging ability was slightly worse than that of VC;The total organisms in the pulp and peel of soft jujube macaques.Thereducing power of the alkali compound is in the range of 50-150 μg/m L,and it is reluctant with the increase of the mass concentration,and the difference in reducing power between VC and sample is significant.On the whole,the total alkaloids in the pulp and peel of the soft jujube macaque have certain antioxidant activity. |
