Bio-fermentation technology is the perfect cross of modern biology,chemistry and engineering science.As an important part of the national strategy of emerging industries,bio-fermentation technology with its high yield,energy saving,environmental protection advantages,made important contributions in food,industry and other fields.The main source of carbon in the fermentation process is glucose,and too much of its amount will affect the quality of the product.So glucose monitoring is very important in the fermentation process.The traditional detection method is a kind of off-line measurement.In which Fermentation broth is taken from the fermenter,and then do the detection of glucose concentration.The traditional detection method will not only waste time but also lead to the wrong glucose concentration,because of the continuous breathing of the bacteria.So the on-line measurement of glucose is necessary.This paper analyzes the enzyme-injectable glucose on-line sensor analyzer which has been developed for the fermentation industry by our laboratory,puts forward its shortcomings and then the design of second-generation online enzyme-injectable glucose analyzer was carried out.(1)Research on the detection method of enzyme injection glucose biosensors based on temperatureThe results of Enzyme-injected glucose biosensor are susceptible to temperature,which can lead to inaccurate measurements.In order to solve this problem,a temperature-based glucose concentration detection method was proposed.By combining the Arrhenius formula in thermodynamics with the kinetic method based on the mechanism model,a new method for the determination of the concentration of glucose in the measured liquid is proposed by measuring the temperature of the reaction mixture and the initial slope of the reaction.The temperature-based detection method can reduce the error of temperature introduction,improve the accuracy of the test results,and can omit the conventional manual calibration steps,simplify the operation,to achieve better on-line detection.(2)Design of the hardware and software system for the second-generation enzyme-injected glucose on-line analyzerThe flow structure and hardware structure for the enzyme-injected glucose on-line analyzer have been designed.The lower computer software system based on STM32 and the hardware system are designed,including three-electrode circuit design,peristaltic pump control circuit design and temperature acquisition circuit design.Functional modules of second-generation online enzyme-injectable glucose analyzer are debugged.And achieve the functions of liquid injection function,three-electrode data acquisition function,temperature measurement function.(3)Carry out test debugging and analysis of the resultsFunctional modules are assembled to obtain a second-generation enzyme-injected glucose analyzer.The post-assembly analyzer is systematically tested,and the test data is analyzed.Finally,the defect and the improved method of the analyzer are presented.
