Objective:To investigate the inhibition of 8-bromine-7-methoxy chrysin(BrMCh R)on Mitogen-activated protein kinase(MAPK)and Nuclear factor(NF-κB)signaling pathway and biological effects in human osteosarcoma MG63 cells activated by IL-1β.To provide a theoretical basis for finding a treatment for osteosarcoma.Methods:Human osteosarcoma MG63 cells was cultured in vitro;Il-1β stimulated the MG63 cells of human osteosarcoma and observed its cellular morphology;Effects of IL-1β activated p-Jnk,p-Erk,p-P38 and IκBα protein expression in human osteosarcoma MG63 cells by Western blot analysis;The effect of IL-1βactivated IL-8,GROα and GROβ gene expression in human osteosarcoma MG63 cells by PCR analysis;Analysis of BrMChR effecting IL-1β activated p-Jnk and p-Erk p-P38 and IκBα protein expression of human osteosarcoma MG63 cells by Western blot;PCR analysis on the effect of BrMChR on IL-1 induced IL-8,GROα and GROβ gene expression in human osteosarcoma MG63 cells.Western blot analysis of BrMChR and its target spot in the MAPK and NF-κB signaling pathway.Results:1.IL-1β had no significant effect on the morphology of human osteosarcoma MG63 cells.2.IL-1β could activate the MAPK and NF-κB signaling pathway in human osteosarcoma MG63 cells,and the expression of p-Jnk,p-Erk,p-P38 protein was significantly up-regulated(P<0.05),IκBα protein expression was significantly down-regulated(P<0.05).3.IL-1βcan effectively reduce the expression of GROα,GROβand IL-8 oncogene in human osteosarcoma MG63 cells(P<0.05).4.BrMChR can significantly inhibit IL-1β activated MAPK and NF-κB signaling pathway,expression of p-Jnk,p-Erk,p-P38 protein significantly down-regulated(P<0.05),IκBα protein expression was significantly up-regulated(P<0.05).5.BrMChRcan reduce IL-1β activate the expression of GROα,GROβand IL-8 genes in osteosarcoma MG63 cells(P < 0.05).6.The effect site of BrMChR on MAPK and NF-κB is TRAF6.Conclusion:1.IL-1β could activate MAPK and NF-κB signaling pathway.2.BrMChR could effectively inhibit MAPK and NF-κB signaling pathwayactivated by IL-1β3.BrMChR inhibited site was TRAF6 which upstream of MAPK and NF-κB pathways.
