Objective:The non-SMC condensin Ⅰ complex subunit G(NCAPG)that organizes the coiling topology of individual chromatids,represents an overexpressed antigen in various cancers and also contributes to restructuring chromatin into rod-shaped mitotic chromosomes and ensuring the segregation of sister chromatid during the cell division.We investigated the relationship between NCAPG expression and the biological behavior in hepatocellular carcinoma to further explore the potential of NCAPG as a therapeutic target.Methods:The expression of NCAPG of tumor samples was detected by Immunohistochemistry.The effect of NCAPG on the cell cycle,apoptosis,proliferation and metastasis was investigated by Western Blot,CCK8,flow cytometry,Transwell assays.Results:The effect of NCAPG on the cell cycle,apoptosis,proliferation and metastasis was investigated by various assays.The expression of NCAPG in five human HCC cell lines(SMMC-7721,HepG2,MHCC-97 H,Hu7 and MHCC-LM3)were higher than in the normal control LO2 cell line,NCAPG was overexpressed in hepatocellular carcinoma compared with the adjacent normal tissue(P < 1/1000),and high levels of NCAPG expression were significantly correlated with recurrence,the time of recurrence,metastasis,differentiation,and TNM stage.Furthermore,elevated NCAPG expression was associated with a poor overall survival(P < 1/20).In addition,the in vitro study further confirmed the ex vivo data;i.e.,knockdown of NCAPG expression reduced hepatocellular cancer cell viability but induced apoptosis,arrested cells at the S phase of the cell cycle and inhibited proliferation.Knockdown of NCAPG expression also inhibited tumor cell migration and invasion capacity in vitro.At the protein level,knockdown of NCAPG expression upregulated Bax,Cleaved Caspase-3,and E-cadherin but downregulated cyclinA1,cdk2,Bcl-2,N-cadherin,and HOXB9.Conclusion:The expression of NCAPG in the tissues of patients with hepatocellular carcinoma was significantly higher than that in the adjacent tissues,which was statistically significant.There is a significant correlation between the expression of NCAPG in hepatocellular carcinoma patients and tumor TNM stage,pathological differentiation,metastasis,recurrence and recurrence time.the relapse free survival time of HCC patients with positive expression of NCAPG in tumor tissue were shorter than negative patients.knockdown of NCAPG expression arrested cell cycle,induced apoptosis,inhibited proliferation and suppressed tumor cell epithelial–mesenchymal transition(EMT).
