Effects Of Acupoint Injection Therapy And Traditional Chinese Medicine Tea Of Treating Disease For The Level Of SIRT 1 Of Castrated Female Rats

Purposes:To observe the effect ofacupoint injection therapy and traditional Chinese medicine tea medicine on the levels of endocrine and SIRT1 of Castrated female rat,in order to explore the mechanism of the treatment of climacteric syndrome and delaying the aging of menopause.Methods: Selected 156 three-month old female Sprague-Dawley(SD)rats,Body weight(250g±20g),in order to more accurately simulate the process of human menopausal ovariande cline,the rats were divided into Sham operation groupand the group of the three groups: partial resection groups set,subtotal resection groups set and totalre section groups set(in each groups set,include modeling group,traditional Chinese medicine tea group,acupoint injection therapy group,traditional Chinese medicine tea and acupoint injection therapy group),a total of 13 groups,12 rats each group.Partial resection group set: Remove the total right ovary,and 50% theleft ovary;Subtotal resection group set: Remove the total right ovary,and 80% the left ovary.Total resection group set: Ovariectomy through ligation.After model establishment,traditional Chinese medicine tea group drinks medicine freely,acupoint injection therapy group chooses acupoints “Guanyuan,Guilai(dou ble),Ganshu(double),Shenshu(double)”to inject Danhong injection with 0.02 ml each acupoint,once a week,tatal 8 times.Applying ELISA method to examine levels of estradiol(E2),follicle-stimulating hormone(FSH)in serum and SIRT1 protein of heart tissue of female rats,and dyeing and observe morphology an d structure of uterine tissue.Results: Compared with the control group,SIRT1 protein and E2 in heart tissue of rat serum in model group were significantly lower,serum FSH levels were significantly higher,The differences were statistically significant(P<0.05);compared with the model group,the treatment group can improve the SIRT1 protein E2 in serum and heart tissue level(P<0.05),lower serum level of FSH(P<0.05);Among them,the levels of E2 and SIRT1 protein were highest in secondary cut traditional Chinese medicine tea+acupoint i njection group,and lowest in FSH level.The morphology of uterine tissue show ed that each treatment group could increase the thickness of endometrial epithe lium,and the stroma was dense,glandular and vascular,among them,the secon dary cut traditional Chinese medicine tea+acupoint injection group treatment effect is the best.Conclusions:(1)In this study,we used three methods of castration,subtotal hysterectomy and total excision to make a successful modelof the rat model of menopause,and simulates the process of human climacteric.(2)Acupoint injection and Chinese herbal tea can slow the aging process inovariec tomized rats,its mechanism may be acupoint injection and Chinese herbal tea can improve the serum E2 in ovariectomized rats and the level of SIRT1 protein in cardiac tissue,decrease the level of serum FSH.(3)the statistical data of each treatment group showed thatIn the process of the gradual decline of the ovary,the early combination therapy is more effective.
