Eggplant as solanaceae nightshade,has hundreds of years of cultivation history in our country,which has rich resources and genetic diversity,provide abundant raw material for breeding improved varieties.The research of genetic relationship analysis of eggplant germplasm resources and genetic diversity,in favor of preservation of eggplant germplasm resources collection and utilization.Therefore,this study using SSR molecular marker and SRAP markers to genetic diversity analysis of 80 of eggplant germplasm resources,and the close relatives of 12 materials for south root knot nematodes,bacterial wilt and verticillium wilt resistance identification,choose strong comprehensive resistance as root stock for grafting,the good situation of comprehensive evaluation of the root stock by investigating the growth potential,fruit quality and yield of tomato.The results are as follows:(1)Using SSR and SRAP molecular marker to collect 80 eggplant resources,screening for 20 SSR primers,amplify the specific band 99,the polymorphic bands was 81.15%;21 of SRAP primers had amplified 207 specific bands,specificity band account for 89.60%.(2)Using the selected primers for 80 material to make SSR molecular marker analysis,and combining with morphological traits to comprehensive analysis of 80 copies of materials.Using NTSYS calculate similarity coefficient variation range of 0.5~1.0,by SSR clustering analysis,when the similarity coefficient was 0.6104,80 material can be divided into seven groups.Based on SRAP amplification product,calculate the similarity coefficient variation range of 0.50~0.91,by SRAP clustering analysis,the similarity coefficient of 0.578,80 eggplant resources can be divided into eight groups.The S.totrvunm and S.totrvum varieties are separated out at first in two kinds of molecular markers,the S.integrifolium Poir.was not separated from Solanum melongena,this shows that the S.integrifolium genetic relationship for the Solanum melongena was closer than S.torvum Molecular markers clustering results with morphological classification,source of fruit shape and has a certain relevance,but not perfectly.(3)Through the related analysis,selection and cultivation of close relationship between 1 S.torvum S101,4S.integrifolium Poir.7of Solanum melongena in total of 12 varieties of materials,to identified the South Root knot nematodes and the resistance of bacterial wilt and Verticillium Wilt,the results are as follows:S.torvum for three kinds of diseases were showed a strong resistance,the S.integrifolium Poir.and the Solanum melongena to Root knot nematode resistance is weak,S.integrifolium Poir.to bacterial wilt for high performance and to Verticillium Wilt resistance for performance differences;Cultivation of tomato bacterial wilt and Verticillium Wilt performance differences.The stronger resistance to overall varieties were solanum torvumS101,Solanum melongena 177,S.integrifolium Poir.166 and y11.(4)Choose the stronger resistance to 177,166 and yll as stock material with Guihong2(small fruit)and Aibili(large fruit)of tomato for grafting.Based on the tomato growth potential,fruit characters and yield in the investigation after grafting,the result shows:the eggplant root stock was root development,the tomato growth potential increased significantly in the middle and later period after grafting;After The fruit hardness of big fruit and small fruit had increased after grafting;Large fruit tomato with eggplant root stock for grafting had significantly increased the fruit soluble solids,soluble sugar content and sugar acid ratio.
