| Hydrogen peroxide is a common reactive oxygen species,mainly produced in cell mitochondria.Hydrogen peroxide is widely used in bleach and disinfectant,the necessary product in biological and physiological process.On the other hand,hydrogen peroxide plays an important role in the process of cell signal transduction and organism immunity,so the detection of hydrogen peroxide in the cell is of great significance.Existing small molecular probes detect hydrogen peroxide by reacting directly to hydrogen peroxide,the identified groups are mainly boric acid derivatives,however,when its reaction with hydrogen peroxide is vulnerable to other oxide interference,can’t monitor the actual content of hydrogen peroxide.In order to solve this problem,we use the reaction of chloride ions can be oxidized into hypochlorite under the catalysis of MPO,join a reverse reaction,designed and synthesized a kind of probe which can specific response to hypochlorite.The probe choose BODIPY as fluorophore,BODIPY has excellent optical performance.The choice of recognition group is phenothiazine,the fluorescence is quenched by the PET of S in phenothiazine and BODIPY.After the probe react with hypochlorous acid,the PET disappeared and probe fluorescence recovery.Fluorescence performance before and after reaction in the form of off-on,so as to achieve specific detection of hypochlorous acid and is not affected by other oxidants.When the chloride ion is oxidized to hypochloric acid,hypochlorous acid can react with the probe,then hypochlorous acid to be restored to the chloride ion,the chloride ion can react with hydrogen peroxide to produce hypochlorous acid again.So between chlorine ions and hypochlorous acid to form the complete cycle,and in the process of the cycle,with the probe reaction is hypochlorite,but actual consumption is hydrogen peroxide,thus probe can effectively be used to monitor the change of the hydrogen peroxide content in cells.The monitoring can provide better tracking disease caused by hydrogen peroxide content change state,has a great help in understanding pathogenesis. |
