Surface Plasmon Resonance (SPR), a traditional biological detection technology with novel characteristics and application, featured by high sensitivity, small-volume sample, good repeatability and stability, marker free and rapid detection, has been widely applied in chemical and pharmaceutical, bio-detection, medical testing. However, current mature SPR detection devices are too huge and expensive, miniaturization is one of the main trends of SPR device research.Based on current research of miniaturized SPR devices, this thesis is focused on the new type and solution of a miniaturized and portable SPR system for near infrared measurement. It gives an overview of SPR sensor technology, principles, trends, as well as mature SPR instruments and miniaturized instrument features. According to the study of many relative research results, a Kretschmann type SPR device is designed with a new optical path and prism structures making a new structure of the reflective prism SPR system. According to the modeling and simulation for the Surface Plasmon Resonance, several key parameters such as the incident angle, prism material, the thickness of golden film has been determined and optimized in this new structure. And design the reflective prism, sensor chips, flow cell, fixtures of prism, as well as light transmitter and receiver. Integrate the transmitter and receiver, reducing the system size, enhancing portability.The device is prototyped and tested with some special experiments, main wavelength range from750to1000nm with broad spectrum detection range from visible region to near infrared zone. System detection sensitivity is up to1.29×10-5RIU at near infrared zone. According to the experiments it stands that the reflective structure has very good stability and repeatability, high sensitivity. It is a compact, portable device with high stability and a very broad spectrum detection range.
