| Objective:To discuss whether gonadectomy can lead to the urodynamics change of the bladder detrusor of female rat; whether gonadectomy can lead to the change of female rat’s bladder histology and the trait of the change. Whether gonadectomy can lead to some active molecule in bladder tissue. Such as the change of the gene and the protein of BK channels, aquaporins (AQPs), acid-sensing ion channel protein3 (ASIC3), whether these changes have something to do with the change of urodynamics. To study the change of the expression of BK channels, aquaporins (AQPs), acid-sensing ion channel protein3 (ASIC3) in rat’s bladder tissue and the relationship with bladder detrusor. To know the relationship between BK channels, aquaporins (AQPs), acid-sensing ion channel protein3 (ASIC3) and urinary tract stimulation. To provide new research idea for clinical urinary tract stimulation and the treatment of OAB.Methods:Choose some adult female rats, build rats’estrogen lack model by giving bilateral oophorectomy and supplying xenoestrogen. The situation of grouping is as follows: dividing all the rats randomly into control group, gonadectomy group and nilestriol group. After the rats were allowed to acclimate for 6 days, give bilateral ovary exploratory surgery to control group, give ovariectomy to the rats in the other two groups. Give bladder fistulization to all the rats, and immobilize the end of fistula to the necks of the rats, feed the rats for using after awaking. Drench nilestriol suspension 1mL/100g to the nilestriol group, once a week, for 4 weeks. Gavage isopyknic normal saline in the rest time. Gavage normal saline 1mL/100g to control group and gonadectomy group, once a day, for 4 weeks.Do urodynamics detection to the three groups’ rats. Take the bladder tissue of the three groups’ rats, give pathology examination. To measure the difference of the content of BK channels, aquaporins (AQPs), acid-sensing ion channel protein3 (ASIC3) by the way of immunohistochemistry stain, RT-PCR and western-blot.Results:Urodynamics hints:In control group, there isn’t obvious instability shrink in storage and urination period. In gonadectomy group, there is instability shrink in storage period, and compare with the control group, the urination interval shorten markedly (P<0.01), the urination times increased markedly (P<0.01). The nilestriol group has few times of instability shrinks compared with gonadectomy group, and has fewer times of urination and longer urination interval (P<0.05)Pathology detection hints that the rats’ bladder mucosa has been destroyed; RT-PCR and western-blot hint:compared with control group, in gonadectomy group, on the bladder the BK channels expression decreased markedly (P<0.01). ASIC3 expression increased markedly (P<0.01). AQPs hasn’t change markedly. In nilestriol group, BK channels expression is higher than that in gonadectomy group (P<0.05) and lower than that in control group (P<0.05). ASIC3 expression is higher than that in control group (P<0.05) and lower than that in gonadectomy group (P<0.05)Conclusion:1. Gonadectomy can lead to the instability shrinks of female rats’ bladder detrusor, it can be remitted by supplying xenoestrogen.2. Castration can lead to female rats bladder mucosal histological changes, these changes may is the pathological basis of the urodynamic changes.3. The gene and the protein expression of BK channels on gonadectomy female rats are lower than that on normal rats, they can rise after supplying xenoestrogen, but can not reach normal level.4. The gene and the protein expression of ASIC3 on gonadectomy female rats are higher than that on normal rats, they can reduce after supplying xenoestrogen, but can not reach normal level.5. In female rats’ bladders, the reducing of BK channels expression and the rising of ASIC3 expression closely related with bladder detrusor hyperreflexia.6. Xenoestrogen may become potential medicine to treat postmenopausal women’s bladder detrusor hyperreflexia. |
