A Correlation Study Of Clinical Characteristics, Drug Efficacies And MicroRNA Plasma Levels In Patients With Generalized Anxiety Disorders

Object:To examine the expression levels of pre- and post-treatment plasma mi R-34 c, mi R-381, mi R-451 in generalized anxiety disorder(GAD) patients and their relation to clinical characteristics and drug efficacies in order to provide basis for revealing the action of micro RNAs in GAD.Methods : 42 patients with GADand 40 healthy controls were enrolled. Basic demographics, first-episode age, total course and current course of research group and generalizeddemographic data of control were added up. The patients were treated with Paroxetine for 12 weeks. Hamilton Anxiety Scale(HAMA) was used to assess the efficacy of Paroxetine and plasma mi R-34 c, mi R-381 and mi R-451 expression levels were detected by real-time PCR.Results:1. The plasma level micro RNA in the patients differed significantly from the control group before and after treatment. mi R-34c(t=-16.96,-5.37, P<0.01)、mi R-378(t=-11.01,-9.71, P < 0.01) and mi R-451(t=-9.80,-6.35, P < 0.01)alldecreasedsignificantly.2. Pre- treatment mi R-34 c, mi R-378 and mi R-451 expression levels were significantly negatively related to the total score of the HAMA(R=-0.57,-0.52,-0.42, P<0.01).Pre- treatment mi R-34 c, mi R-378 expression levels were significantly negatively related to the mental anxiety score of the HAMA(R=-0.52,-0.41, P<0.01).3. After treatment plasma expression level of mi R-34c(t=-9.90, P<0.01)elevatedcompared with pretreatment in research group.4. Pre- and post-treatment mi R-34 c, mi R-378 and mi R-451 expression levels were significantly negatively related to thetotal course( R=-0.84,-0.83,-0.71,-0.68,-0.59,-0.58, P<0.01).mi R-451 expression level was negatively related to the first-episode age in research group(R=-0.43,-0.58, P<0.01).Conclusion1. It is possible for mi R-34 c, mi R-378 and mi R-451 to take part in the pathogenesis of GADs2. The plasma mi R-34 c level in GAD patients is affected by the treatment of Paroxetine.3. The severer anxiety degree is, the lower the plasma mi R-34 c, mi R-378 and mi R-451 expression level becomes.The plasma mi R-34 c, mi R-378 expression levels may be related to the mental anxiety symptoms.4. The longer total course is, the lower the plasma mi R-34 c, mi R-378 and mi R-451 expression level becomes. These mi RNAs may be potential indications of refractoriness of GAD.
