Chapter1:In this chapter, the composition, basic principles and characteristics of SPR biosensor were described. The SPR analysis methods and the applications in detection of food, detection of environment, pharmaceutical research and life sciences were introduced in detail. The concept, selection and advantages of aptamer were outlined briefly. In addition, some immobilized methods of aptamer and applications in electrochemical sensors, optical sensors and piezoelectric sensors were described as well.Chapter2:A surface plasmon resonance (SPR) assay had been developed for the detection of lysozyme. In this protocol, lysozyme aptamer instead of traditional antibodies was immobilized to a sensor chip Au by self-assembled method. The experimental parameters were optimized in term of injection time, flow rate and pH. As a result, The linear range of the assay for lysozyme was1μmol/L~100μmol/L and a detection limit of0.5μmol/L was obtained.the aptasensor also presents high specificity for lysozyme, which was not affected by other proteins.Chapter3:The sensor chip was modified with1,4-benzenedithiol by self-assembled method. The gold nanoparticles was absorbed onto the dithiol-modified Au sensor chip. Then the interaction of lysozyme aptamer and lysozyme was investigated by SPR technology. As a result, the developed sensing system showed a linear response to concentration of lysozyme range from0.2μmol/L to20μmol/L. The detection limit of lysozyme was determined to be0.09μmol/L. The SPR response enhanced by using the modified Au sensor chip.Chapter4:A novel aptamer-based biosensor combined with gold nanoparticles amplification for the determination of kanamycin with SPR technology was presented. In order to optimize the experimental conditions, a series of measurements including injection time and pH were researched in this work. The direct assay and modified assay were investigated for real-time kanamycin detection. The results showed the modified assay could play a part in enhancing SPR signal.
