| This research was carried out to investigate the effects of different dose of daidzein on laying performance in laying hens and the safety evaluation involved. Six hundred and forty56-wk-old Hyline Brown were randomly assigned to4groups with8replicates of20birds each. Fed diets supplemented with0(control),10mg,50mg and100mg of daidzein/kg for12wk. Through the feeding trials, slaughter performance measurement and immunohistochemistry, we investigated the effects of daidzein on laying performance in laying hens. Through the RT-PCR technology we investigated the.expression of related genes. The research provided theoretical basis for feed production enterprise and farms (households) to use of daidzein scientifically. The results show that:1. In the whole trial period, feed intake was not significantly effected by daidzein (P>0.05). Supplementation with50mg of daidzein/kg in the diet for four weeks significantly improved the laying rate of eggs, egg mass and the efficient feed conversion (P<0.05). Compared with50mg of daidzein/kg, supplementation with100mg of daidzein/kg significantly reduced the laying performance (P<0.05). The level of estradiol and progesterone in resum were not significantly effected by daidzein (P>0.05).2. There was significantly difference in the index of oviduct (P<0.05). The index of ovary, uterus, small yellow follicle and the length of oviduct, the number of small yellow follicle, large white follicle were not significantly effected by daidzein (P>0.05).3. Compared with group I, there were no significantly differences in the morphology of ovary and uterus. Interstitial cell structure is normal, there were different development stages of follicles. The mucosa cells, muscle cells and serosa layer cells were relatively complete. For immunohistochemical analysis, we statisticed the positive cell number of IL-1β,and no significantly differences(P>0.05).4. The expression quantity of FSHR in ovary base organization was significantly effected by daidzein(P<0.05), and the groupâ…¢ got the highest expression. Compared with groupIII, the expression quantity of FSHRã€LHRã€P450arom were decreased significantly(P>0.05). The expression quantity of P450arom in theca cell layer and LHR in granular cells layer were no significantly difference(P>0.05). The expression quantity of IL-1β and TNF-α mRNA of ovary tissue were not significantly effected by daidzein (P>0.05). |
