| Natural products are important resources for the development of new drugs, which is the basis of many clinical drugs for synthesizing natural products or semi-synthetic. So far, microorganism is still an important source of lead compounds. Fungi are important groups of microorganisms. They exist from fresh water to salt water, from the surface of plants to the inside plant tissues, from terrestrial to marine. That immune suppressants, anti-pathogenic fungal agents and cholesterol-lowering agents from fungal antibiotics have made tremendous contributions to human disease in clinical resistance. Fungi are metabolites manufacturing plant with unique biological activity and novel structure.In this thesis, four fungal material were selected including two plants endophytic fungi and two marine fungi. Their crude extract from solid fermentation was isolated with the use of Nuclear Magnetic Resonance, UV, IR spectroscopy, mass spectrometry and so on to identify compounds.35compounds were isolated and identified which9ones were new. Bioactivities of these new compounds and other compounds were evaluated.Three compounds were isolated and elucidated from the strain Fusarium oxysporum F01820cultured in PDA solid fermentation medium including one fusaric acid (ZHQ-01-065-03), one phenylalanine-like compound (ZHQ-01-058-04) and one steroidal compound (ZHQ-01-059-01).Twenty-five compounds were isolated and elucidated from the strain Pycnidiophora dispersa F01510cultured in PDA solid fermentation medium with half of sea water.There were9new compounds including ZHQ-01-100-01, ZHQ-01-103-01,ZHQ-01-105-01,ZHQ-01-118-01,ZHQ-01-119-01,ZHQ-01-122-01, ZHQ-01-122-02, ZHQ-01-125-01,ZHQ-01-128-01.Three major series of compounds were isolated including four sulochrin and derivatives, four asterric acid and derivatives, nine depsidone and derivatives. In addition, there are two sterols, a sphingolipid compound, a benzene dicarboxylic acid compound, a new compound of soluchirin binding with asterric acid, and a5-hydroxy-3-methoxybenzoic acid, one hamavellone B, one curvulapyrone. For the the new compounds, there is no significant antitumor and antibacterial activity.Two compounds were isolated and elucidated from the strain Neosartorya stramenia MCCC3A00273cultured in PDA solid fermentation medium with half of sea water. ZHQ-02-009-04is a cytochalasin compound, ZHQ-02-009-05is a known steroid.Five compounds were isolated and elucidated from the strain Tritirachium sp. MCCC3A00152cultured in PDA solid fermentation medium with half of sea water including ZHQ-02-024-02, ZHQ-02-030-01, ZHQ-02-030-02, ZHQ-02-033-03, ZHQ-02-039-01.The structure of ZHQ-02-033-03and ZHQ-02-039-01is similar to xanthoquinodin. They both show cytotoxic activity on human cervical cancer Hela cells and human malignant pleural mesothelioma ACC-MESO-1cells.In conclusion, this work indicated that fungi are the important source of natural products with many kinds of structure types and diverse bioactivities including antimicrobial and antitumor. |
