Objective:To investigate promoter methylation status and expression of ferroportin1(FP1) in the substantia nigra of unilateral6-OHDA-treated rat model with Parkinson Disease.Methods:6-OHDA was injected into rat unilateral intramedial forebrain bundle(MFB).The numbers of TH (tyrosine hydroxylase) immunoreactive neurons were examined by immunohistochemical methods;the expression of FP1mRNA and the promoter methylation status of FP1were detected by RT-PCR and MSP respectively.Results:Compared with unlesioned side and control group,the number of TH immunoreactive neurons in the6-OHDA injected side was reduced obviously (twithin=20.619, P<0.01; tbetween=18.404, P<0.01),FP1mRNA expression was down-regulated(twithin=8.123,P<0.01; tbetween=7.609, P<0.01); Methylation of FP1gene promoter in experimental group increased significantly compared with control group(X2=20,P<0.01).Conclusion:Unilateral injection of6-OHDA into rat intramedial forebrain bundle leads to down-regulation of FP1mRNA expression; Unilateral injection of6-OHDA into rat intramedial forebrain bundle leads to hypermethylation of FP1DNA promoter region, which may have a relationship with down-regulation of FP1mRNA expression.
