| Objective:To create the animal model with acute pancreatitis(AP) which can adapt to scientific research,and to investigate the influence of Dachaihu Tang on pancreatic external secretion in rats with acute pancreatitis.Methods:Experiment1:24Wistar rats were divided into3groups randomly,including normal group,model group and treatment group.Each group was8rats.2h before the inducti-on of AP model,the group C was given Dachaihu Tang(1ml/100g).AP animal models was induced by subcutaneous injection of eaerulein (50μg/kg·h) on rats of group B and group C, and normal group, by subcutaneous injection of saline50ug/(kg·h),six times consecutively.Immediately by the induction of the models, the treament group was given Dachaihu Tang(1ml/100g) orally while group A and group B were given saline in the same way.6h later,bile pancreatic juice was collected to detecte Ca2+density, amylase and lipase of rats in each group.Experiment2:30Wistar rats were divided into three groups randomly:group A(normal group),group B(model group),group C(treatment group).Each group was10rats.2h before the induction of AP model,the group C was given Dachaihu Tang(1ml/100g).AP animal models was induced by subcutaneous injection of caerulein (50ug/kg·h) on rats of group B and group C, and group A, by subcutaneous injection of saline50ug/(kg·h),six times consecutively. Immediately by the induction of the models.the treament group was given Dachaihu Tang(1ml/100g) orally,group A and group B were given saline in the same way.6h later, pancreatic tissue of rats in each group was stained and then the pancreatic acinar Ca2+-Mg2+ATPase and calcium concentration was detected in order to investigate the effect of Dachaihu Tang on calcium pupm and Ca2+concentration in acinar cells.Results:1.Compared with normal group,amylase of pancreatic juice in rats of model group was significantly higerer,while the treament group was lower than model group(P<0.05).Ca2+in pancreatic juice of model group was significantly lower than that of normal group and the two groups were significant difference (P<0.05).2. Compared with group C,Ca2+concentration in pancreatic acinar cells of group B was significantly higher, the two groups were significant difference.(P<0.05).3. The changes of Ca2+-Mg2+ATPase in Group A. group B and group C:light mic-roscopy showed different shades of brown particles distributed on the cellular membrane,cytoplasm and nuclear membrane of pancreatic acinar cells. Most amount of slices of normal control group showed positive,only small amount showed negat-ive while most amount of sliced slices of AP group showed negative.And the treament group showed more positive than that of group B.Group B and group C were significant difference(P<0.05).Conclusion:Ca2+-Mg2+ATPase in pancreatic acinar cells of rats with AP was decreased significantly while Ca2+concentration was increased.Dachaihu Tang may reduce the Ca2+concentration in the acinar cells by the increase of Ca2+-Mg2+ATPase which can decrease cell calcium overload in the process of AP,and thus affect the pancreatic exocrine secretion function. |
