This study used experimental analysis of SSR markers validation on23major maize varieties andinbred lines in Hebei province from the period of2005to2011. It aims to:(i) Improve the DNA extraction method: exploring the rapid extraction method to single seed ofmaize, which can increase the purity of OD260/OD280between1.8-2.0;(ii) Optimize PCR amplification system: making use of the current equipments to optimize thePCR amplification system to a better effect on the basis of the laboratory conditions foroptimization.(iii) Build a standard sample map: selecting23standardized samples with good market fromHebei province in recent years to establish fingerprint database to validate the maize hybrids andthe parental genetic authenticity, and its technology application system.Based on the experiment, fingerprints DNA bands data of20primer sites were acquired throughthe detection to20pairs of the basic core primers, and finally found that:(i) Differences loci varieties≥2, judged to be different species;(ii) Differences loci varieties=1, judged as highly similar ones;(iii) Species differences loci=0, judged to be the suspect with the variety.Currently, the corn DNA fingerprint technology has been widely used in a variety Genuinenessand validation. According to incomplete statistics from Maize Research Center of BeijingAcademy of Agricultural Sciences, there are several hundred new approved maize varieties andthousand varieties participating into countries and provinces testing, of which more than400national trial varieties, and600varieties have been issued variety rights have made use of thistechnology to make identification. Through this, a lot of disputes due to the varieties ofauthenticity and variety rights have been solved. Now the seed management departments, such asBeijing, Sichuan etc have taken this technology.
