| With corn starch as material, this paper discussed the preparation process of indigestible dextrin and its influence on blood glucose lipid and antioxidant level. The reactant of indigestible dextrin was identified with common corn starch as material, HC1as activator and Termanyl120L Type L as enzyme.Preparation process of indigestible dextrin has been optimized. The addition of acid was8%. The temperature of preparation was160℃. The time of preparation was50min. The addition of enzyme was0.5%. The time of alcohol-precipitation was1h. Under this condition, we could obtain high-purity indigestible dextrin after vacuum freeze drying, and its purity was87.2%which was determined by enzyme-HPLC method.The determined method of indigestible dextrin molecular weight by HPLC was founded. The condition of chromatogram:mobile fhase is three-braize-water, velocity of flow is0.8mL/min, sample size is20μL, shromatographic is OHpak SB-803, and column temperature is30℃. The molecular weight of indigestible dextrin before and after alcohol-precipitation were as followed:before the alcohol-precipitation was2835, after the alcohol-precipitation was4067. At the same time, the new established method has been study by the precision, reproducibility, stability and accuracy.The result of physical and chemical characteristic of indigestible dextrin show that the cation-exchange capacity was0.3056mmol/g, the water holding capacity was10.22g/g, and the solubility was70g/30ml. The viscosity of the solution of indigestible dextrin was increased with the increasing of concentration and decreased with the increasing of temperature and shear rate. But with the increase of pH, the number of freezing and salts concentration did not changed significantly.Using indigestible dextrin as animal experiments, we study the GI of indigestible dextrin and influence of indigestible dextrin on blood glucose lipid and antioxidant level. Result showed that the GI of indigestible dextrin was17belonging to the food of low GI. The normal mice which taking indigestible dextrin grew well and no adverse reactions. Indigestible dextrin treatment diabetic mice improved behavior and lowered the blood glucose evidently. The concentration of TG and TC in serum were decreased. The concentration of HDL-C was increased. In serum and live, the concentration of MDA was decreased, and the activities of GSH-PX and SOD were increased. |
