To purifications and chatacterization the activity composition of analgesia from earthworm. To research the effect of the activity composition to analgesia:1. Five fractions(A-E) were isilated from earthworm by CM-Sephadex C-50 column with step-wise elution.Among them two fractions(C,D) are determined neurotoxic on isolated rat phrenic nerve-diaphragm preparetions.Four fractions(C1,C2,D1,D2) were isilated by Sephadex G-75 column. SDS-PAGE analysis of C1,D1,D2:The molecular weight of C1 is 28.3kD, the molecular weight of D1 is 26.4~32.3kD, and the D2 is 26.0-31.8kD.2. The effect to pain threshold:the activity composition of analgesia from earthworm was determined using the hot plate test and the acetic acid writhing test. The activity composition exhibited a dose-dependent analgesic action in mice as determined with hot-plate test and acetic acid writhing test. The significant analgesia of activity composition was seen 30min to 60min after its administration. Earthworms extract on the influence of pain threshold is proportional to the measurement. The model of rat nerve-hemidiaphram preparation have been made.Twitchs were elicited by stimulate delectrically through nerve(NS) and diaphragm(MS) alternately with supramaximal voltage at a rate of 2 shocks per minute by rectangular-wave pulse of about 10ms duration.C1 is determined neurotoxic on isolated rat phrenic nerve-diaphragm preparetions.3. The effect of earthworm on wound healing in scalded rats:The model of deep partial thickness burn was established and wound healing time, use the activity composition of analgesia from earthworm to treat the model.The activity composition from earthworm can shorten the epithelium cicatrization time and reduce inflammation.
