Objective:Application of ultrafine research in the field of Traditional Chinese Medicine is one of the hotspots. In this paper, take Notopterygium and Radix Saposhnikoviae as the research objects, study on the preparation of ultrafine powder, analyse the physical/chemical properties and the efficacy of ultrafine powder, comparative with fine, provide experimental evidences for the applicability of ultrafine grinding technology of on TCM which containing volatile oil, heat-sensitive and polysaccharides.Methods:(1) Study on the preparation process:The particle size distribution of ultrafine powder and all levels of ultrafine powder were measured by the BT-9300H laser particle sizer to screen the crushing process. (2)Reserch of physical properties:Investigated in ultrafine microstructure, surface area, mobility, bulk density and moisture absorption and so on, and the difference between powders. (3)Reserch of chemical property.The changes of active ingredients of fine and ultrafine powder were compared by TLC, HPLC and IR. (4)Leaching experiment:select the ferulic acid as an indicator and select the chromone as an indicator, comparison of the leaching rate of powder and superfine active ingredients at different times. (5) The study of efficacy:Taking Notopterygium as the research object, study antipyretic and analgesic efficacies of fine and ultrafine powder.Results:(1) Notopterygium and Radix Saposhnikoviae ultrafine powder were prepared in lower temperature by vibration mill. (2)There is little difference of physical property between fine and ultrafine powder of Notopterygium and Radix Saposhnikoviae. (3)The results of TLC, HPLC and IR indicate that ultrafine comminution not change the active ingredients of medicine, the loss of active ingredients can be avoided by Cryogenic Vibration Mill. (4) The leaching effect of Notopterygiumul traf ine powder is much better than fine; leaching rate of accumulation of Radix Saposhnikoviae ultrafine powder already over 70% at 10min, Radix Saposhnikoviae ultrafine powder, show no significant difference between them. (5)The results of mouse acetic acid writhing test show that, ultrafine powder and fine of Notopterygium can significantly inhibit the writhing response in mice, Analgesia of ultrafine powder is significantly better than fine. Compared with the control group, the group of fine and the group of ultrafine powder both had significant antipyretic effect, but there was no significant difference of the antipyretic effect between the group of fine and the group of ultrafine powder.Conclusion:The study indicates that Cryogenic Vibration Mill can be used for Notopterygium and Radix Saposhnikoviae ultrafine powder, ultrafine comminution has little effect on the physical property of medicine, more dissolution of active ingredients, increased efficacy of antipyretic and analgesic of Notopterygium.
