| Limonium bicolor Kunze(Bge.), commonly named as broomy grass and so on similarity the grass drought weeds in China. The full plant is traditional drug, to heal bleeding, cancer nephritis and other weakness symptom.The thesis is aimed to develop herbal resources of Shannxi through researching the dried buds and flowers of Limonium bicolor Kunze(Bge.). This thesis has a systematic study focused on the polysaccharide extraction, purification process including removing protein and the decolorization process. The antioxidant activity of polysaccharide is tested with preliminary analysis its structure.(1) polysaccharide extractionLimonium bicolor Kunze(Bge.) polysaccharide was obtained by using hot water extraction and ultrasonic wave extraction. On the basis of one-factor tests,the method of response surface analysis(RSA) was carried out to determine the optimal condition of the extraction. The using hot water optimum extraction conditions were as follows:83℃,1.8h, 16.2 (water/dry weight),2 times. The extract rate of polysaccharide is 1.731% based on the above mentioned conditions. The ultrasonic wave extraction optimal condition were:ultrasonic extraction power 85W,85℃,61min,11 (water/dry weight),2 times.Under the optimized condition,the extraction rate of polysaccharide was 1.879%. The result shows that the ultrasonic method is time-saving,lower temperature and higher the extraction rate of polysaccharide,compared with using hot water. However, the polysaccharide structure may be changed by using of ultrasonic wave extraction.(2) Process of removing proteinBy comparing the four methods of protein removal including by trichloroacetic acid, Sevage method, papain,papain-Sevage, papain-Sevage method was choosed as the optimal method finally. The optimum conditions were substrate concentration 2% of papain, pH 7.0, temperature 75℃, hydrolysis time 2h, repeat 3 times with Sevags method. The amount of protein removed was 83.72% while polysaccharide remained 80.25% during the deproteinization process.(3) Process of decolorizationContrasting four decolorization methods such as hydrogen peroxide oxidation method, live carbon attaching method, reverse micelle solution method and macroporous rosin adsorption method, it was hydrogen peroxide oxidation method that had the most effect in decolorizing polysaccharide. The optimum conditions were pH 8.5, temperature 55℃, hydrolysis time 2h,1.5. The amount of decolorization was 87.75% while polysaccharide remained 87.97% during the decolorization process. (4) Study the antioxidant activity of Limonium bicolor Kunze(Bge.) polysaccharidesThe results showed that Limonium bicolor Kunze(Bge.) polysaccharides had the cleaning capability of superoxide radical,hydroxy radical and DPPH.Especially the superoxide radical, and hydroxy radical had satisfied scavenging effect. |
