This thesis includes three chapters:Chapter 1: Recent progress on myriapoda mitochondrial genome and genetic variation analysis of Otostigmns (O.) politus populationsChapter 2: Replenishment of the mitochondrial genome of Scolopendra multilan. Mitochondrial DNA extraction, long-PCR amplification and primer-walking sequencing methods were carried out for determining the complete mt DNA sequence of Scolopendra multilan.(1) The complete mitochondrial genome of Scolopendra multilan is 13,734 bp in size. Its components, gene order, gene orientation, low percentage of guanine and high percentage of thymine as well as skew of base GC and AT are more similar with myriapodas.(2) It is found many adjacent genes overlaps.(3) There are only 2 standard stop codes TAA and TAG for protein genes, this is different from other myriapodas.(4) Compara to Lithobins forficatus, there is only one tRNA differernce in rearrangement.Chapter 3: We explored genetic variation analysis of using part of mitonchondrial cob gene sequence of Otostigmus(O.) politus. A total of 26 haplotypes were defined by sequnces variance, and 58 individuals were collected from 11 locations in five populations. Phylogenetic trees(ML and Baysian) of haplotypes are very confusion. The diagrams of TCS and MSN reveal that Otostigmus(O.) politus have many ancestal haplotypes (HB1, SHY1, SYC1, SYC2, SJZ1), which have many radiation forming a "star-like" shape. Haplotype diversityis(h) between 0~1.000 and nucletid diversity (π) between 0~0.18162, which shows that the range of genetic diversty is very large. According to molecular clock of Arthropods, our data suggests that the distribution pattern of Otostigmus(O.) politus is very original, which formed after the middle of Tertiary Period, so it maybe not influenced by glacial periods in the Pleistocene. Therefore this is why there are many ancestal haplotypes. It is populated that South China wild boar and Central China wild boar are original populations, but Ussurian Ussurian wild boar and North China wild boar are modern populations, which is embodiment of annimals how to suit to climatic change.
