Buckwheat is one kind of buckwheat crop with high nutritional value, which is rich in high quality proteins, starch, dietary fiber, mineral elements, vitamins, flavonoids and other bioactive substances. Among them, buckwheat protein has the potential of medicinal value. As a new type of phytoprotein resources, the research of buckwheat protein has attracted people’s much attention in recent years.In this study, the application of Osborne method to extract protein, and calculated the extraction rate of the four proteins. To study the inhibitory activity of protein hydrolysate on angiotensin converting enzyme. The main results of this study are as follows.1. This experiment extract four kinds of protein in Buckwheat (albumin, globulin, gliadin and glutenin) by using sequential extraction methods. The experiment measured the total protein content in skimmed buckwheat was 12.7g per 100g skim. The proportion of the four proteins were albumin 38.98%, globulin 12.00%, gliadin 0.47%, glutenin 20.47%. The sum of the four protein content is 9.14g, the protein extraction rate was 71.97%.2. Through the selection test of protease, the enzymatic hydrolysate of albumin has inhibitory activity on ACE only when it under the action of neutral protease and compound protease. The enzymatic hydrolysate of glutenin has inhibitory activity on ACE only when it under the action of neutral protease, acidic protease and protease K. The other two proteins did not show ACE inhibitory activity. Under the action of neutral protease, the inhibition rate of ACE was the highest, and the inhibition rate were:albumin 33.38%, glutenin 34.87%.3. Through single factor and orthogonal experiment, we determined that under the action of neutral protease, the optimum enzymolysis conditions of Albumin were:substrate concentration 5.0%, enzyme concentration 5.0%, enzyme hydrolysis temperature 40℃, enzymatic hydrolysis time 120min. Under the condition of the optimum enzymatic hydrolysis, the inhibition rate of ACE was 36.04%; The optimum enzymolysis conditions of glutenin were:substrate concentration 4.0%, enzyme concentration 6.0%, enzyme hydrolysis temperature 40℃, enzymatic hydrolysis time 60min. Under the condition of the optimum enzymatic hydrolysis, the inhibition rate of ACE was 35.6%.
