Based on the previous study about the whole-cell bio-detection and bio-adsorption, this project aims at using the synthetic biology technology(eg."BioBrick") to integrate the gold ion selective detection based on GolS regulator with the gold ion selective adsorption based on the surface display of GolB protein, and by improving the original resistance to the toxicity of the gold ion in the ion probe of the whole cell, the integration will finally achieve a integration of expression system in gold ions detection and adsorption that is regulated by the gradients of gold ions in environment, which makes success in selective detection and adsorption of gold ion from the mixture of several ions. Then, it would be achievable for B.subtilis to selectively detect and absorb a trace of gold ions by homologous recombination of this system, which provides an important direction for other biological detection and biological reparations of toxic heavy metal ions. At the same time, this study will make this kind of construction of whole cell detection and adsorption more patterned, then realize a more convenient way of gold ions detection and adsorption.
