| Polymerase chain reaction (PCR) technology is a kind of DNA amplification technique outside the body, which has been widely applied to biomedicine and other related fields. At present, the vast majority of PCR reaction is carried out in the traditional commercial-PCR, which uses larger metals to control thermal cycling temperature. Because metal has big heat capacity, the elevation and decrease speed of temperature is slower. A complete PCR cycle usually requires 1h~2h and even longer period of time. In addition, capillary electrophoresis analysis of micro-fluidic detection system has been able to complet in a smaller volume chip. If the continuous flow PCR chip (CF-PCR) and capillary electrophoresis analysis chip are integrated together, biochemical analysis will achieve integration and miniaturization. Therefore, this paper dasigns CF-PCR chip system based on MCU and FPGA, which mainly studys on PCR chip production,amplification and temperature control system.This paper fabricates a CF-PCR chip with the microelectronic technique and micromechanical process by using laboratory pre-designed CF-PCR chip technology territory, including a micro flow-through chip of glass material, and three silicon chips consist of Pt membrane micro-heaters and temperature sensors. The intelligence temperature control system is designed based on MCU and FPGA platform, which separates the temperature control algorithm from MCU by using FPGA to achieve. This design not only displays many advantages of FPGA, such as high integration,reliability well,designing flexibility,faster computing speed and so on, but alse greatly improves the temperature control precision. Among them, the MCU mainly realizes functions of AD sampling control, keyboard keys control and transmitting data directive to FPGA; FPGA mainly realizes functions of temperature curve fitting, PID control algorithm, as well as PWM waveform generator. With the combination of external hardware circuit, this system completes a multi-channel temperature signals in the acquisition,conversion,display and other functions. This system effectively realizates the three temperature thermostat control.By using CF -PCR chip system, plant DNA sample is amplified and the PCR product is detected by means of agarose gel electrophoresis. The experimental results show that this system can basically realize the amplifition of DNA samples. |
