| Objective To investigate the association between AKT1 gene SNP and major depressive disorder.Methods All of the subjects were recruited from the Han population in Chongqing.taking the method of a case-control study (case-control study), into a clear diagnosis of 127 cases of patients with MDD and 100 cases of healthy adults. and taking the method of conventional phenol - chloroform extraction of whole blood genomic DNA samples, then choosing the polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) method ,Analysing AKT1 gene 5 single nucleotide polymorphism( SNP) sites between the two groups in the gene frequency, genotype frequency , haplotype and linkage disequilibrium test (linkage disequilibrium test, LDT) .Result (1) AKT1 gene SNP3 (rs3730358) C allele frequencies for the 92%, T allele frequency was 8% in MDDAND C allele frequency of 97%, T allele frequency of 3 % in the control group , the differences between the two groups was significant (p=0.024),MDD group SNP3 genotype frequency (C/C=84%,C/T =16%), the control group (C/C=94%,C/T=6%), the differences between the two groups are statistically significance (p = 0.020). No difference between the two groups of the gene frequency and genotype frequency of the remaining four SNP loci.(2) linkage disequilibrium test (LDT) results show that, SNP3/SNP4/SNP5 close linkage (D' for the 0.965~0.997), SNP1/SNP2/SNP3 moderate linkage (D' for 0.622 ~ 0.720). haplotype analysis of these sets of SNP loci showed that, CGG haplotyp(eχ~2 = 5.592, p = 0.018, OR = 0.634,95% CI = 0.434 ~ 0.926) and the TGG haplotype (χ~2 = 4.712, p = 0.029, OR = 2.901,95% CI = 1.065 ~ 7.907) of the SNP3/SNP4/SNP5,There were difference between the MDD group and the control group.ATC haplotype(χ2 = 4.707, p = 0.030, OR = 6.216,95% CI = 0.949 ~ 40.714)of the SNP1/SNP2/SNP3,the two groups also have significant differencesConclusion (1) AKT1 gene SNP3 (rs3730358) with MDD is associated(2) AKT1 gene SNP1/SNP2/SNP3. and SNP3/SNP4/SNP5 linkage disequilibrium (3) Haplotype of Certain AKT1 gene with MDD is associated(4) AKT1 gene could be a susceptibility gene of theMDD... |
