| Objective: Ropivacaine is a new part-anaesthesia drug. It is revealed that 1% ropivacaine would injure the ultrastructure of spinal cord and nerve root early(after six hours) in CSEA. In order to explore the potentional cell damage and the alterations of nitric oxide(NO), nitric oxide synthase (NOS) in brain stem , hippocampi tissues and serum after celiac injection ropivacaine in wistar rats and provide some experimental data for the assessment of the security of ropivacaine .Methods: Seventy-two male Wistar rats were randomly divided into 4 groups of 18 animals each in group. The animals were randomized to receive normal saline, 0.5%, 0.75% or 1.0% ropivavaine 0.1ml every 100g body weight of Wistar rats. After the celiac injection administration 6h,12h and 24h, all rats were drawed blood approximately 2.5ml from femoral artery, 0.5ml blood (heparinized) to determine DNA damage of lymphocyte cell. 2.0ml blood to determine the content of NO and the activity of NOS. We rapidly distill homogenate of brain stem and hippocampi to determine the content of NO and the activity of NOS. Using the Alkaline single cell gel electrophoresis (SCGE) assay to observe the potential damage of the cells which exposed to diffirent concentration of ropivavaine.Results: (1) The NO levels of hippocampi and brain stem nerve cell in 0.5%,0.75% and 1% ropivavaine group were higher than that in normal saline in diffirent time phase, while only in 1.0% ropivavaine group the NO levels were significantly dissimilar compared with normal saline group (P<0.05). (2) The nitric oxide synthase (NOS) activity of hippocampi and brain stem nerve cell in 0.5%,0.75% and 1% ropivavaine group were higher than that in normal saline in diffirent time phase, while only in 1.0% ropivavaine group the NO levels were significantly dissimilar compared with normal saline group (P<0.05).(3) The comet tail ratio and tail length of brain stem, hippocampi and lymphocyte cell were steady during observation in normal saline, 0.5%, 0.75% ropivavaine group, while in 1.0% ropivavaine group the levels were dissimilar compared with normal groups after celiac injection 6h (P< 0.05). After celiac injection 12h and 24h, the lever of that in every experimental groups were higher than that in normal saline group, while only in 1.0% ropivavaine group the result was significantly dissimilar compared with normal saline group (P<0.05).Conclusions: (1) There isn't obviously effect of 0.5% and 0.75% ropivavaine on rats' brain stem and hippocampi After celiac injection 24h . (2) 1% Ropivavaine is damaging to the rats' brain stem , hippocampi and blood lymphocyte cell, which lead to the increasing of comet tail ratio and tail length. |
