| Background and ObjectiveBlastocystis hominis (B.hominis) is a more and more interesting intestinal parasit protozoan. It has been ignored after its discovery by Alexief in 1911 because of various morphological feature, difficultion in detection etc. There are few study on B. hominis in China. Although a few specialist have worked out some articles on pathology, morphology, immunity and physiology. Most of the study are at preliminary stage and more detailed research is necessary.In view of special biochemical characteristic of B.hominis , proposed life cycle, we study the morphology of B.hominis by special or universal stain methods through light microscopy of B.hominis and electronic microscopy. From the result of own morphological study we can even infer certain organism function. Then infective the mouse whose immunity is normal or low by various quantities. Last we created the experiment model of the Blastocystis hominis disease. Materials and Methods 1.Observation of MorphologyIn present study, the B.hominis positive stool specimen was used to inoculate Locker's biphasic egg medium with 10% bovine sera supplement and modified RPMI-1640 culture medium(RPMI-1640). This culture was incubated at 37C. The parasites should be transfer every 48-72h to assure the viability. At the same time, antibiotics, penicillin and streptomycin, were added to the new culture. Afterward, the liquid culture was extracted every day to be stained with Iodine and H.E, respectively. |
