| The Camellia sinensis is one of the major cash crops, and the Tea industry take an important place. In farming it takes 3-5 years to reach its reproductive development until die. It takes about year and a half from budding phase to fruit setting stage and this process consumption a lot of nutrient which lower the products of the Camellia sinensis leaves.EMBRYONIC FLOWER(EMF) genes maintain vegetative development by repressing the flowering program, LEAFY and P450 genes known as promoting plant flowering. Camellia sinensis(Tie guanyin) was taken as the material for research in this article. Genes mentioned above involved in flowering regulation pathway were cloned by RT-PCR and RACE. The results were as follows:1 Cloning of the full length DNA of EMBRYONIC FLOWER2(EMF2) by RT-PCR and RACEThe full-length cDNA of Camellia sinensis EMF2 is 2145bp nucleotides long. It contains an ORF encoding 622 amino acids(Mw=157318kDa, pI=4.89 ),a 5′untranslated region of 196bp and a 3′flanking sequence of 278bp. Compared with EMF2 gene of others plants by Blast software, Camellia sinensis EMF2 cDNA usually had 83 % , 81 %, 79 % , 76 % and 75 % identity with that of Vitis vinifera, Lycopersicon enculentum, Ricinus communis, Silene latifolia and Populus trichocaprpa EMF2 genes, respectively. This cDNA had been registered in GenBank with accession number GQ477137.2 Cloning of cDNA frament of LEAFY(LFY) by RT-PCR and RACEA 670bp cDNA fragment was obtained. Compared with LEAFY of other plants, It showed that the deduced Camellia sinensis LEAFY gene had high similarity with others plants LEAFYSs by Blast software. Camellia sinensis LEAFY cDNA usually had 99 % identity with that of Magnolia virginiana,Paulownia tomentosa,Mimus ringens,Buddleja dvidii,Mimulus kelloggii and Verbena officinalis EMF2 genes.3 Cloning of cDNA frament of P450 by RT-PCR and RACEA 772bp cDNA fragment was obtained. Compared with P450 of other plants, It showed that the deduced Camellia sinensis P450 gene had high similarity with others plants P450s by Blast software. Camellia sinensis P450 cDNA usually had 79 %,79 %,73 %identity with that of Populus trichocarpa,Arabidopsis thaliana,Ricinus communis P450 genes, respectively.To sum up, EMF2, LEAFY, P450 genes plays an important role in the growth of Camellia sinensis, its also on the control of the Camellia sinensis. May this research of clone these genes above set a theoretical foundation for the future. |
