| Scutellaria barbata D.Don as one of the Chinese traditional medicines has wide applications and multi-functions. Recently studies showed that it has good antitumor function. It's commonly and viably to improve the officinal factors by induction of polyploidy. In this study, we induce and identify polyploidy according to this theoretical support. At same time, in vitro propagation system was developped. At last, Total flavonoid, one of officinal factors, was determined.The stem tips, as explants, were treated by 0.2% NaClO solution for 5 minites. Vacuum seedlings grew up in MS medium, 1cm stem tips and 0.8cm stem segments were chosen as explants few times late.In vitro propagation system was developped: 1cm stem tips were chosen as the explants, MS + 6-BA 0.5 mg·L-1 + IBA 0.1 mg·L-1 was an optimal medium for the bud differentiation; its multiplication coefficient is 6.5. 1cm stem tips were chosen as the explants, MS + 6-BA 0.5 mg·L-1 + NAA 0 mg·L-1 was an optimal medium for the bud differentiation; its multiplication coefficient is 6.1. 0.8cm stem segments were chosen as the explants, MS + 6-BA 0.5 mg·L-1 + NAA 0 mg·L-1 was an optimal medium for the bud differentiation; its multiplication coefficient is 5.7. 0.8cm stem segments were chosen as the explants, MS + 6-BA 0.5 mg·L-1 + NAA 0 mg·L-1 was an optimal medium for the bud differentiation; its multiplication coefficient is 4.8. Therefore, MS + 6-BA 0.5mg-L"' + IBA 0.1mg·L-1 was an optimal medium for the bud differentiation.Calli induction: Explants was death in MS+6-BA+NAA medium, both leaves and stem segments were chosen, can't be differentiated.Polyploidys induction: The highest induction rate of Scutellaria barbata D.Don polyploidys is 10% while stem tips were treated by 0.2% colchincine soluction for 3 hours, the survial rate is 25%.Comparisons of normals and polyploidys from morphology to microhisology, polyploidys were determined by flow cytometer at last. Three plant lines were chosen from four plant lines what were identified random. Their contents of total flavonoid were 1.88%,1.85% and 1.69% orderly, non-treated plant line was 1.67%. Recovery rate of total flavonoid was 99.1%, standard deviation (RSD) is 3.56%. |
