Water is one of key factors which restrict larvae-pupal development of overwintered Asion corn borer Ostrinia furnacalis (Guenée). It plays an important role in exploring the effects of water intake on the pupal development to study development-related protein of Asion corn borer after drinking water. Plasma is temporary metabolize place in Asion corn borer, which is charged with many important physiological function. The mechanism in water drinking for Asian corn borer larvae can be efficiently explored while its protein changed. The overwintered lavea of Asion corn borer were chosen as the research material and the activities of the three protective enzymes (catalase, CAT; superoxide dismutase, SOD and peroxidase, POD) were surveyed in the larvae of Asion corn borer before and after drinking water. Three protective enzymes study was just on the start. By using comparative proteomic technology some differentially expression protein were indentified in Asion corn borer before and after drinking water. The indentified protein were classified according to the functional of them. In this thesis, the works which we did are showed as follows:The activities of the three protective enzymes (catalase, CAT; superoxide dismutase, SOD and peroxidase, POD) were surveyed in the larvae of Asion corn borer before and after drinking water.The results showed that the activities of the three protective enzymes were lower in the body of overwintered Asion corn borer before drinking water,but significantly increased from the first day to the fourth day after drinking water. SOD activity increased to highest in the first day after drinking water, and it kept a constant level from the frist to the third day. CAT activity increased to highest in the second day, and POD increased to highest in the third day. The activities of the three protective enzymes respectly decreased to lower levels in the fourth day. It suggested that there was a very close relationship between the activities of the three protective enzymes and development metabolism of overwintered larvae after drinking water.The plasma was full of lots of proteins which having important functions in the control of insect metamorphosis development. So comparative proteomic technology two-dimensional gel electrophoresis in conjunction with high-throughput MALDI-TOF-TOF MS were employed to investigate protein differentially expression patterns of overwintered larvae during larvae-pupal metamorphosis. 13 protein spots were differentially expressed in the first day with 9 proteins sports were detected to be upregulated more than 3-fold, while 4 of them were downregulated more than 3-fold respectly. Compared with the first day, there were 21 protein spots significantly changed in the forth day after drinking, and 11 protein spots of them increased in expression abundance with 10 protein spots decreased differently. 18 proteins were identified by MALDI-TOF-TOF MS successfully, including Paramyosin,acyl-coenzyme A oxidase,Chitinase-like protein Idgf1,cGMP-specific 3',5'-cyclic phosphodiesterase,ommochrome binding protein,Serine/threonine-protein kinase ATM,Tyrosine-protein phosphatase,Serine/threonine-protein phosphatase 2B catalytic subunit 1,Collagen alpha-1(XIII) chain,Negative elongation factor E,ATP-dependent RNA helicase vasa and unknowed proteins.These identified proteins which classified by the NCBI and UniporkKB databases and related references, belonged to 6 functional categories, including immunity, metabolism, cell structure, signal transduction, development, transcription and traslation.The newly established 2-DE refrence map of plasma proteins may serve as an important tool for further understanding of the protein function which differently expressed in time during larvae-pupal metamorphosis of overwintered Asion corn borer after drinking water.
